柔肝通络汤对脑缺血再灌注损伤大鼠Bcl-2蛋白表达的影响

摘　　要：目的探讨柔肝通络汤对脑缺血再灌注损伤大鼠Bcl-2蛋白表达的影响。方法将78只SD大鼠随机分为5组:假手术组、模型组及柔肝通络汤高、中、低剂量组。假手术组及模型组连续1周灌胃蒸馏水。柔肝通络汤各剂量组分别给予高、中、低剂量的柔肝通络汤灌胃,最后一次灌胃30 min后,制成大脑中动脉栓塞模型,保持缺血状态2 h后,分别按再灌注3、12、24 h 3个时间点取脑。取脑前评估大鼠肢体神经功能缺损程度,取大鼠梗死侧脑组织,进行HE染色检测观察梗死脑组织细胞形态,并用蛋白免疫组化染色检测Bcl-2蛋白。结果与假手术组大鼠相比,模型组大鼠神经功能缺损程度评分升高(P<0.01),梗死侧神经元细胞损伤加重,Bcl-2蛋白表达显著升高(P<0.01)。与模型组比较,柔肝通络汤各组神经功能缺损程度评分下降(P<0.05),大鼠梗死侧神经元细胞损伤情况明显减轻,Bcl-2蛋白表达均显著升高(P<0.01)。结论柔肝通络汤能有效改善大鼠神经功能障碍,减轻缺血再灌注后细胞损伤,增加Bcl-2蛋白表达,起到脑保护作用。Objective:To explore the effect of Rougan Tongluo Decoction on the expression of Bcl-2 protein in rats with cerebral ischemia reperfusion injury.Methods:A total of 78 SD rats were randomly divided into 5 group:sham operation group,model control group and Rougan Tongluo Decoctiongroups with high,medium and low dose.Distilled water or high,medium and low concentrations of Rougan Tongluo Decoction were given by gavage for 7 d continuously.After 30 min of the last gavage,the middle cerebral artery embolization model was established;after ischemia for 2 h,the brain was taken at 3 time points of reperfusion 3,12 and 24 h,respectively.Before taking the brain,the rats were evaluated for the neurological deficit degrees;HE staining and protein immunohistochemical staining were performed on rat brain tissue after taking the brain.Results:Compared with the sham group,the neurological deficit scores in the model group were increased(P<0.01),the neuronal cell injury in the infarcted side was aggravated,and the expression of Bcl-2 protein was significantly increased(P<0.01).Compared with the model group,the neurological deficit scores in each of Rougan Tongluo Decoction group was decreased(P<0.05),the neuronal cell injury in the infarcted side of rats was significantly reduced,and the expression of Bcl-2 protein was significantly increased(P<0.01).Conclusion:Rougan Tongluo Decoction can effectively improve rat nerve dysfunction,reduce cell injury after ischemia reperfusion,increase the expression of Bcl-2 protein,and play a role in brain protection.
