摘　　要：目的探讨PYCR1基因在肝细胞癌(hepatocellular carcinoma,HCC)中的表达及其与临床病理特征和预后的关系。方法采用qRT⁃PCR和Western blot技术对80例HCC与癌旁组织的PYCR1 mRNA和蛋白表达水平进行检测。采用Cox比例风险模型对影响HCC患者预后的因素进行单因素、多因素分析。采用Kaplan⁃Meier和Log⁃rank法进行预后分析。结果HCC组中PYCR1 mRNA和蛋白相对表达量均明显高于癌旁对照组(均P<0.05)。HCC组织中PYCR1表达水平与肿瘤大小、卫星灶、血管侵犯、TNM分期有关(均P<0.05)。PYCR1表达水平是影响HCC患者5年总体生存率和无瘤生存率的独立因素(均P<0.05)。HCC组织中PYCR1低表达组患者5年总体生存率、无瘤生存率均明显高于PYCR1高表达组(均P<0.05)。结论PYCR1在HCC组织中高表达,与肿瘤大小、卫星灶、血管侵犯、TNM分期有关。组织中PYCR1表达水平与HCC患者预后关系密切。Objective To explore the expression of PYCR1 gene in hepatocellular carcinoma(HCC)and its relationship with clinicopathological features and prognosis.Methods Expression levels of PYCR1 mRNA and protein in HCC and adjacent tissues from 80 patients were detected by qRT⁃PCR and Western blot.Cox proportional hazards model was used to analyze the prognostic factors of HCC patients.Kaplan⁃Meier method and the log⁃rank test were used to analyze the prognosis.Results Relative expressions of PYCR1 mRNA and protein in HCC group were higher than that in adjacent tissue group(P<0.05 for both comparisons).Expression of PYCR1 was correlated with tumor size,satellite lesions,vascular invasion and TNM stage(P<0.05 for all comparisons).PYCR1 expres⁃sion level was an independent factor for affecting 5⁃year overall survival rate and disease⁃free survival rate in HCC patients(P<0.05 for both comparisons).The 5⁃year overall survival rate and disease⁃free survival rate in patients with low expression of PYCR1 in HCC tissues were greater than those in patients with high expression of PYCR1(P<0.05 for both comparisons).Conclusions PYCR1 is highly expressed in HCC tissues,which is related with tumor size,satellite lesions,vascular invasion,and TNM stage.Expression of PYCR1 is closely related with the prognosis of HCC patients.
