摘  要:目的探讨E-4031对遗传性长QT综合征人类ether-a-go-go相关基因(h ERG)通道功能的影响。方法构建杂合型WT/G572R-h ERG细胞株,根据E-4031是否干预分为观察组和对照组。Western blot检测两组h ERG通道蛋白表达,全细胞膜片钳技术检测两组h ERG通道激活电流、尾电流、稳态失活电流和失活恢复电流的变化。结果观察组155-k Da蛋白质条带明显高于对照组,差异有统计学意义(P<0.05)。观察组h ERG通道激活电流和尾电流的最大电流幅度[(416.32±90.37)、(897.25±92.05)p A]均高于对照组[(219.34±28.98)、(566.61±121.60)p A],差异均有统计学意义(均P<0.05);观察组和对照组h ERG通道稳态失活电流半失活最大电压[(-83.39±1.45)m V比(-69.05±2.58)m V]差异有统计学意义(P<0.05)。结论E-4031能改善WT/G572R-h ERG通道功能。Objective To investigate the effect of E-4031 on human ether-a-go-go-related gene(h ERG)channel function in hereditary long QT syndrome.Methods Heterozygous WT/G572 R-h ERG cell lines were constructed and divided into observation group and control group according to whether incubated with E-4031.Western Blot was performed to detect the expression of h ERG channel.The whole-cell patch-clamp technique was used to assess active current of h ERG,tail current,steady-state inactivation current and recovery current.Results The 155-k Da protein band was significantly higher in observation group than in control group(P<0.05).The activation current of the h ERG channel and the maximum tail current were significantly higher in observation group[(416.32±90.37),(897.25±92.05)p A]than in control group[(219.34±28.98),(566.61±121.60)p A](all P<0.05).The maximum half-inactivation potential of steady-state inactivation of h ERG current was significantly different between the two groups[(-83.39±1.45)m V vs(-69.05±2.58)m V](P<0.05).Conclusion E-4031 may improve WT/G572 R-h ERG channel function.
