摘 要:目的探讨表没食子儿茶素没食子酸酯对寒潮诱发高血压大鼠脑卒中的作用影响。方法健康雄性SD大鼠分为3组,将其中的寒潮诱发高血压大鼠脑卒中模型分为-模型组与药物组,每组各15只。模型组给予双蒸水灌胃,药物组给予表没食子儿茶素没食子酸酯灌胃。正常组16只不建模,给予双蒸水灌胃。治疗2周与4周后,记录3组大鼠的逃避潜伏期、神经行为学评分、收缩压;治疗4周后,比较3组大鼠脑组织的糖皮质激素诱导蛋白激酶1(SGK1)和蛋白激酶B(Akt)蛋白相对表达水平。结果治疗2周与4周后,模型组[(88.09±11.23)、(88.87±11.23)s]与药物组[(67.24±7.22)、(55.20±11.76)s]的逃避潜伏期均显著长于正常组[(43.98±8.11)、(44.88±3.34)s],药物组则较模型组显著缩短,差异均有统计学意义(P<0.05)。治疗2周与4周后,模型组[(2.56±0.18)、(2.38±0.14)分]与药物组[(1.50±0.11)、(1.24±0.12)分]的神经行为学评分均显著高于正常组[(0.44±0.03)、(0.43±0.02)分],药物组则较模型组显著降低,差异均有统计学意义(P<0.05)。治疗2周与4周后,模型组[(178.09±12.09)、(179.09±13.44)mmHg]与药物组[(155.92±13.11)、(143.98±12.76)mmHg]的收缩压均显著高于正常组[(122.49±13.19)、(123.00±10.48)mmHg],药物组则较模型组显著降低,差异均有统计学意义(P<0.05)。治疗4周后,模型组(6.22±0.44、5.82±0.41)与药物组(3.29±0.29、2.81±0.33)大鼠脑组织的SGK1和Akt蛋白相对表达水平低于正常组(0.92±0.12、0.67±0.09),药物组低于模型组,差异均具有统计学意义(P<0.05)。结论表没食子儿茶素没食子酸酯在寒潮诱发高血压大鼠脑卒中模型中的应用能促进降低血压,改善大鼠的记忆能力与神经行为能力,抑制SGK1、Akt蛋白表达。Objective To investigate the effects of epigallocatechin gallate on stroke induced by cold wave in hypertensive rats.Methods Healthy male SD rats were divided into three groups.The cold wave induced hypertensive rat stroke model were divided into model group and drug group,each group 15 rats.The model group were given double distilled water treatment,and the drug group were given epigallocatechin gallate treatment.There were no model were established in the normal group(n=16),and were also given double distilled water treatment.After 2 and 4 weeks of treatment,the escape latency,neurobehavioral scores,and systolic blood pressure of the 3 groups of rats were recorded;after 4 weeks of treatment,the brain tissues of the 3 groups of rats were compared with glucocorticoid-induced protein kinase 1(SGK1)and protein Relative expression level of kinase B(Akt)protein.Results After 2 and 4 weeks of treatment,the escape latency of the model group[(88.09±11.23),(88.87±11.23)s]and the drug group[(67.24±7.22),(55.20±11.76)s]were significantly longer than the normal group[(43.98±8.11),(44.88±3.34)s],the drug group were significantly shorter than the model group,and the differences were statistically significant(P<0.05).After 2 and 4 weeks of treatment,the neurobehavioral scores of the model group[(2.56±0.18),(2.38±0.14)points]and the drug group[(1.50±0.11),(1.24±0.12)points]were significantly higher than in the normal group[(0.44±0.03),(0.43±0.02)points],the drug group were significantly lower than the model group,and the differences were statistically significant(P<0.05).After 2 weeks and 4 weeks of treatment,the systolic blood pressures of the model group[(178.09±12.09),(179.09±13.44)mmHg]and the drug group[(155.92±13.11),(143.98±12.76)mmHg]were significantly higher than those in the normal group[(122.49±13.19),(123.00±10.48)mmHg],the drug group were significantly lower than the model group,the differences were statistically significant(P<0.05).After 4 weeks of treatment,the relative expression levels of SGK1
