摘 要:目的探讨COX-2基因rs5275多态性是否会影响慢性精神分裂症的发病,并进一步检测是否影响慢性精神分裂症患者的阴性、阳性等精神症状。方法本研究于2014~2016年收集564例慢性精神分裂症受试者及419例健康对照志愿者。采用CRF(Case report form)表收集所有受试者的一般社会人口学和临床资料。采用阳性、阴性症状量表(Positive and negative syndrome scale,PANSS)对慢性精神分裂症患者的精神症状进行评测。采用TaqMan SNP方法对COX-2基因rs5275多态性进行基因分型为CC、CT、TT。比较两组rs5275等位基因和基因型分布频率的差异,并分析基因型与慢性精神分裂症患者临床精神症状之间的关系。结果在健康对照组和慢性精神分裂症组之间,COX-2基因rs5275位点的等位基因、基因型频率分布比较,差异无统计学意义(χ^2=1.33,df=1,P=0.25;χ^2=1.45,df=2,P=0.49)。按COX-2基因rs5275位点的基因型对慢性精神分裂症组进行分组分析,患者阳性症状在组间存在显著差异(P=0.02)。结论COX-2基因rs5275位点可能不影响慢性精神分裂症的发病机制,但可能影响慢性精神分裂症患者的阳性症状。Objective To explore whether the COX-2 gene rs5275 polymorphism can affect the onset of chronic schizophrenia and to further detect whether it affects the negative and positive mental symptoms of patients with chronic schizophrenia.Methods A total of 564 chronic schizophrenia subjects and 419 healthy control volunteers from 2014 to 2016 were collected.CRF(Case report form)was used to collect general sociodemographic and clinical data of all subjects.The Positive And Negative Syndrome Scale(PANSS)was used to evaluate the mental symptoms of patients with chronic schizophrenia.The COX-2 gene rs5275 polymorphism was genotyped into the CC,CT,TT using the TaqMan SNP method.The differences in the distribution frequency of rs5275 alleles and genotypes between the two groups were compared.The relationship between genotypes and clinical mental symptoms in patients with chronic schizophrenia was analyzed.Results There was no significant difference in the distribution of alleles and genotype frequencies at the rs5275 locus of the COX-2 gene between the healthy control group and the chronic schizophrenia group(χ^2=1.33,df=1,P=0.25;χ^2=1.45,df=2,P=0.49).The chronic schizophrenia group was analyzed according to the genotype at the rs5275 locus of the COX-2 gene.There were significant differences in the positive symptoms of patients between the groups(P=0.02).Conclusion The rs5275 locus of COX-2 gene may not affect the pathogenesis of chronic schizophrenia,but may affect the positive symptoms of patients with chronic schizophrenia.
