摘　　要：目的建立能同时测定独圣活血片中延胡索乙素、紫堇碱、四氢小檗碱、儿茶素、表儿茶素、美迪紫檀素、香附烯酮和α-香附酮含量的HPLC法。方法采用HPLC法,色谱柱为Waters SunFire C 18(250 mm×4.6 mm,5μm),柱温:30℃;流动相:乙腈-1 mL·L^-1冰醋酸,梯度洗脱;流速:0.9 mL·min^-1;检测波长分别为242 nm(香附烯酮和α-香附酮),280 nm(儿茶素、表儿茶素、美迪紫檀素、延胡索乙素、紫堇碱和四氢小檗碱)。采用SPSS 26.0统计软件对独圣活血片中8种成分的含量进行聚类分析和主成分分析。结果8种成分分别在2.96~59.20,2.42~48.40,1.78~35.60,5.47~109.40,6.29~125.80,0.58~11.60,7.26~145.20,3.58~71.60μg·mL^-1范围内线性关系良好(r≥0.9991);平均回收率分别为98.44%,96.94%,97.26%,99.06%,99.43%,97.28%,100.08%,98.75%,RSD值分别为1.19%,1.26%,1.56%,1.35%,0.75%,1.44%,0.60%,0.93%;10批样品聚类分析为2类。结论该方法操作简便、重复性好,可作为独圣活血片质量控制的科学依据。Objective To establish an HPLC method for the determination of tetrahydropalmatine,corydaline,tetrahydroberberine,(+)-catechin,epicatechin,medicarpin,cyperotundone andα-cyperone in Dusheng Huoxue Tablets simultaneously.Methods The separation was performed by using Waters SunFire C 18(250 mm×4.6 mm,5μm)as the chromatographic column with acetonitrile-1 mL·L^-1 acetic acid solution as the mobile phase in a gradient elution mode.The column temperature was 30℃,and the flow rate was set at 0.9 mL·min^-1.The detection wavelengths were set at 242 nm for cyperotundone andα-cyperone,280 nm for(+)-catechin,epicatechin,medicarpin,tetrahydropalmatine,corydaline and tetrahydroberberine.The cluster analysis and principal component analysis were conducted for the content results of the 8 constituents in Dusheng Huoxue Tablets by SPSS 26.0 statistical software.Results 8 components showed good linear relationships within the ranges of 2.96-59.20,2.42-48.40,1.78-35.60,5.47-109.40,6.29-125.80,0.58-11.60,7.26-145.20 and 3.58-71.60μg·mL^-1(r≥0.9991),whose average recoveries were 98.44%,96.94%,97.26%,99.06%,99.43%,97.28%,100.08%and 98.75%with the RSDs of 1.19%,1.26%,1.56%,1.35%,0.75%,1.44%,0.60%and 0.93%,respectively.The samples of 10 batches of Dusheng Huoxue Tablets could be divided into 2 categories.Conclusion The method is simple with good repeatability.This study lays a foundation for the quality control of Dusheng Huoxue Tablets.
