摘　　要：目的:研究磷酸烯醇丙酮酸羧激酶1(PCK1)在肾透明细胞癌中的表达及其临床意义。方法:采用免疫组织化学法检测30对肾癌组织和癌旁组织,观察比较PCK1在肾癌组织和癌旁组织中的蛋白表达水平差异,并结合癌症基因图谱(TCGA)数据库,分析PCK1的表达量和肾癌患者临床特征和预后的关系。结果:PCK1在癌旁组织中的蛋白表达量明显高于肿瘤组织(P<0.05)。TCGA数据库分析结果显示,PCK1在肾癌中的mRNA表达量明显低于癌组织(P<0.05),且PCK1在低病理分期和无转移患者中的表达量明显高于高病理分期和出现转移的患者(P均<0.05);Kaplan-Meier分析结果显示,PCK1高表达组患者生存率明显高于低表达组(P<0.05);单因素和多因素COX风险回归模型也提示,PCK1高表达患者的死亡风险较低(P<0.05)。结论:PCK1的表达与肾透明细胞癌患者的临床进展呈负相关,有望成为该病的生物标记物。Objective:To investigate the expression of phosphoenolpyruvate carboxykinase 1(PCK1)in clear cell renal cell carcinoma and its clinical significance.Methods:Immunohistochemical approach was used to study and compare the differential expression of PCK1 in 30 paired samples of renal cancer and adjacent tissues.With reference to the Cancer Gene Atlas(TCGA)database,the relationship between the PCK1 expression level,the clinical characteristics and prognosis in patients with renal cancer patients was analyzed.Results:The protein expression of PCK1 in adjacent tissues was significantly higher than that in tumor tissues(P<0.05).Analysis of TCGA database showed that the PCK1 mRNA expression was significantly lower in renal cancer than that in cancer tissues(P<0.05),and higher in patients with low pathological stages or without metastasis than in those with high pathological stages or metastasis.Kaplan-Meier analysis showed that the high PCK1 expression group had significantly higher survival rate than the low PCK1 expression group(P<0.05).Univariate analysis and multivariate COX hazard regression models also indicated that patients with high PCK1 expression had low risk of death(P<0.05).Conclusion:In patients with renal clear cell carcinoma,PCK1 expression negatively correlates with the clinical progression,and therefore can be a promising biomarker of the disease.
