摘  要:[目的]淀粉是植物发育过程中重要的能源物质,也是影响作物品质的重要物质之一。明确谷子淀粉合成调控相关基因对于开展谷子品质分子育种具有重要意义。[方法]本试验对谷子及其它5种禾本科作物直链淀粉合成关键基因Waxy进行聚类,并对其基因特征进行了分析;对Waxy基因在谷子中的表达模式进行分析;以名优谷子品种晋谷21为材料,构建淀粉合成相关基因的共表达网络。[结果]确定了6个物种中Waxy氨基酸序列系统发育关系、基因结构及蛋白基序,明确了Waxy蛋白具有糖基转移酶(GT_1)和淀粉合成酶催化区(GT_5)2个保守性结构域;确定了Si4g02980基因是谷子籽粒淀粉合成的主效基因。WGCNA筛选出含淀粉合成相关基因数目较多的2个模块,确定了一批包括转录因子的候选基因。[结论]本研究结果可为进一步鉴定和克隆谷子淀粉合成调控关键基因及谷子品质分子育种提供理论支撑。[Objective]Starch is an important energy substance in plant development and one of the important substances affect⁃ing crop quality.To identify the genes related to the regulation of starch synthesis in foxtail millet is of great significance for mo⁃lecular breeding to improve foxtail millet quality.[Methods]The waxy gene,a key gene of amylase synthesis in millet and five other gramineous crops,was clustered and its gene characteristics were analyzed.The expression pattern of waxy gene in fox⁃tail millet was analyzed.The co-expression network of starch synthesis related genes was constructed using Jingu21,a famous foxtail millet variety,as materials.[Results]The phylogenetic relationship,gene structure and protein motifs of Waxy amino acid sequences in six species were determined.The results showed that Waxy protein had two conserved domains:glycosyl⁃transferase(GT_1)and starch synthetase catalytic region(GT_5),and Si4g02980 gene was determined to be the the main waxy gene in foxtail millet.WGCNA screened out two modules containing a large number of genes related to starch synthesis and identified a batch of candidate genes including transcription factors.[Conclusion]The results of this study provide theoreti⁃cal support for further identification and cloning of key genes regulating starch synthesis and molecular breeding to improve fox⁃tail millet quality.
