Dectin-1/Syk信号通路介导脂多糖诱导的BV2细胞中的炎症反应

摘　　要：目的探讨脂多糖(LPS)诱导BV2细胞神经炎症反应中树突状细胞相关C型凝集素1(dendritic cell-associated C-type lectin-1,Dectin-1)及其下游脾酪氨酸激酶(spleen tyrosine kinase,Syk)信号通路发挥的作用。方法采用LPS建立BV2细胞炎症模型。将BV2细胞分为对照组、LPS 3 h组、LPS 6 h组、LPS 12 h组和LPS 24 h组,然后用免疫印迹法检测Dectin-1,Syk和p-Syk的表达,随后选出LPS最佳刺激时间。根据LPS最佳刺激时间,再将BV2细胞分为空白对照组、对照+LAM组、对照+PIC组、LPS组、LPS+LAM组和LPS+PIC组;用免疫印迹法检测Dectin-1、p-Syk、肿瘤坏死因子α(TNF-α)和一氧化氮合酶(iNOS)的表达,观察抑制Dectin-1和Syk对促炎因子表达的影响。结果与对照组相比,LPS诱导3,6,12,24 h组Dectin-1,Syk和p-Syk蛋白在BV2细胞中的表达均增加,且在24 h表达量最高(P<0.05)。与空白对照组比较,对照+LAM组和对照+PIC组中各蛋白表达无明显差异(P>0.05);与LPS组相比较,LPS+LAM组Dectin-1,p-Syk,TNF-α和iNOS的蛋白表达明显下调(P<0.05)。类似的,与LPS组比较,LPS+PIC组TNF-α和iNOS的蛋白表达也明显下调(P<0.05)。结论Dectin-1/Syk信号通路可作为治疗神经炎症反应的潜在靶点。Objective To investigate the role of dendritic cell-associated type C lectin 1(Dectin-1)and its downstream spleen tyrosine kinase(Syk)signal pathway in neuroinflammatory response of BV2 cells induced by lipopolysaccharide(LPS).Methods The inflammatory model of BV2 cells was induced by LPS.BV2 cells were divided into control group,LPS 3 h group,LPS 6 h group,LPS 12 h group and LPS 24 h group.The expression of Dectin-1,Syk and p-Syk was detected by Western blotting,and the best stimulation time of LPS was screened.According to the optimal stimulation time of LPS,BV2 cells were divided into blank control group,blank control+LAM group,blank control+PIC group,LPS group,LPS+LAM group and LPS+PIC group.The expression levels of tumor necrosis factor-α(TNF-α),nitric oxide synthase(iNOS),Dectin-1 and p-Syk were detected by Western blotting.Results Compared with control group,the protein expression of Dectin-1,Syk and p-Syk was increased in BV2 cells induced by LPS for 3,6,12,24 h,and the expression was the highest at 24 h(P<0.05).There was no significant difference in the expression of each protein between control group and control+LAM group,control+PIC group(P>0.05).Compared with LPS group,the protein expression of Dectin-1,p-Syk,TNF-αand iNOS was significantly down-regulated in LPS+LAM group(P<0.05).Similarly,compared with LPS group,the protein expression of TNF-αand iNOS was significantly down-regulated in LPS+PIC group(P<0.05).Conclusion Dectin-1/Syk signal pathway can be used as a potential target for the treatment of neuroinflammatory response.
