摘 要:目的:研究富硒黄芪多糖对白血病K562细胞的凋亡诱导作用。方法:采用水提醇沉法制备富硒黄芪多糖。靶细胞选择白血病K562细胞,富硒黄芪多糖作用K562细胞后,采用四甲基偶氮唑蓝(Methyl Thiazolyl Tetrazolium,MTT)比色法检测细胞增殖活性。采用流式细胞术测定K562细胞凋亡率。采用流式细胞术测定Caspase-3活性。结果:标准曲线法测定所提取的黄芪多糖含量。普通黄芪多糖含量:37.90%,富硒黄芪多糖含量:48.70%。富硒黄芪多糖表现出浓度依赖性地抑制K562细胞增殖作用,24 h、48 h和72 h的IC50值分别为376.52μg/mL、283.19μg/mL和197.75μg/mL。200μg/mL和400μg/mL富硒黄芪多糖作用K562细胞48 h后,流式细胞术采用Annexin V/PI双染色方法,结果显示出凋亡细胞含量明显增高,凋亡率分别是13.35%和72.70%,且均以晚期凋亡细胞为主。200μg/mL、400μg/mL富硒黄芪多糖处理K562细胞12 h和24 h后细胞后Caspase-3活性增高到72.54%。结论:富硒黄芪多糖具有诱导白血病K562细胞凋亡作用。Objective: To study the inducting effects of Se-enriched astragalus polysaccharide on apoptosis of leukemia K562 cells. Methods: The Se-enriched astragalus polysaccharide was prepared by water extraction and alcohol precipitation. The target cells were selected from leukemia K562 cells. After Se-enriched astragalus polysaccharides treated on K562 cells, cell proliferation activity was detected by MTT assay. the apoptosis rate of K562 cells were determined by flow cytometry. Caspase-3 activity was determinated by flow cytometry. Results: The content of astragalus polysaccharides was determined by standard curve method. Ordinary astragalus polysaccharide content was 37.90%, and Se-enriched astragalus polysaccharides was 48.70%. The inhibition of K562 cell proliferation in a concentration dependent manner was demonstrated in Se-enriched astragalus polysaccharide. IC50 values were 376.52 μg/mL, 283.19 μg/mL and 197.75 μg/mL in 24 hours, 48 hours and 72 hours respectively. After K562 cells were treated with 100 μg/mL and 400 μg/mL Se-enriched astragalus polysaccharides for 48 hours, the apoptotic cells were significantly increased, the apoptotic rate was 13.35% and 72.70% respectively, most of them were late-stage apoptotic cells. After K562 cells were treated with 100 μg/mL and 400 μg/mL Se-enriched astragalus polysaccharides for 12 hours and 24 hours, the activity of Caspase-3 increased was 72.54%. Conclusion: The Se-enriched astragalus polysaccharide can induce apoptosis of leukemia K562 cells.
