摘  要:目的:探讨lncRNA ZFAS1对宫颈癌细胞放射敏感性的影响以及其作用机制。方法:实验设置pcDNA组、pcDNA-ZFAS1组、si-con组、si-ZFAS1组、IR+si-con组、IR+si-ZFAS1组、si-ZFAS1+anti-miR-con组、si-ZFAS1+anti-miR-193a-3p组、IR+si-ZFAS1+anti-miR-con组、IR+si-ZFAS1+anti-miR-193a-3p组。qRT-PCR检测miR-193a-3p和ZFAS1表达;Western blot检测蛋白表达;流式细胞术检测细胞凋亡;细胞克隆形成实验检测宫颈癌细胞的放射敏感性;双荧光素酶报告实验检测荧光活性。结果:宫颈癌放射抵抗组织和不同剂量射线照射后的宫颈癌细胞中ZFAS1高表达,miR-193a-3p低表达;沉默lncRNA ZFAS1可促进宫颈癌细胞凋亡并增加细胞放射敏感性,促进Cleaved Caspase-3、γ-H2AX表达,抑制Cyclin D1表达。ZFAS1靶向调节miR-193a-3p表达;抑制miR-193a-3p表达逆转了沉默ZFAS1对宫颈癌细胞的放射增敏和细胞凋亡促进作用。结论:lncRNA ZFAS1可增加宫颈癌细胞的放射敏感性,并促细胞凋亡,其机制可能与调控miR-193a-3p有关,为宫颈癌的放疗提供新靶点和新思路。Objective:To investigate effect of lncRNA ZFAS1 on radiosensitivity of cervical cancer cells and its mechanism.Methods:Experimental setup pcDNA group,pcDNA-ZFAS1 group,si-con group,si-ZFAS1 group,IR+si-con group,IR+si-ZFAS1 group,si-ZFAS1+anti-miR-con group,si-ZFAS1+anti-miR-193a-3p group,IR+si-ZFAS1+anti-miR-con group and IR+si-ZFAS1+anti-miR-193a-3p group.Expressions of miR-193a-3p and ZFAS1 were detected by qRT-PCR;protein expression was detected by Western blot;apoptosis was detected by flow cytometry;radiosensitivity of cervical cancer cells was detected by cell clone formation assay;dual luciferase reporter assay was used to detect fluorescence activity.Conclusion:lncRNA ZFAS1 can increase radiosensitivity of cervical cancer cells and promote apoptosis.The mechanism may be related to regulation of miR-193a-3p,which will provide new targets and new ideas for radiotherapy of cervical cancer.
