摘　　要：目的检测长链非编码RNA(lncRNA)序列相似家族83成员A-反义核糖核酸1(FAM83A-AS1)在肺腺癌(LUAD)中的表达水平,探讨lncRNA FAM83A-AS1和LUAD患者临床病理学特征及预后的关系。方法分析2015年1月至12月间80例行外科手术治疗的LUAD患者相关临床资料。采用Real-time PCR方法检测80例LUAD组织,并与癌旁正常肺组织中lncRNA FAM83A-AS1的表达水平进行对照分析。采用卡方检验LUAD患者lncRNA FAM83A-AS1水平与临床病理特征的关系;Kaplan-Meier法分析总生存期(OS);Cox回归模型分析影响LUAD预后的危险因素。结果与肺正常组织相比,LUAD组织中lncRNA FAM83A-AS1表达明显上调(P<0.05)。lncRNA FAM83A-AS1水平升高与肿瘤大小、淋巴结转移和临床TNM分期有关(P<0.05)。lncRNA FAM83A-AS1高表达的LUAD患者总生存期短(p<0.05)。COX生存分析显示,lncRNA FAM83A-AS1基因表达为肺癌预后的危险因素,高表达提示LUAD患者临床预后不良。结论lncRNA FAM83A-AS1在LUAD组织中表达显著升高,且与患者肿瘤大小、淋巴结转移、TNM分期和预后密切相关。Objective This paper aims to detect the expression level of long non-coding RNA FAM83 A-AS1 in lung adenocarcinoma,and to explore its relationship with clinicopathological parameters and prognosis.Methods The clinical data of 80 LUAD patients undergoing surgical treatment between January and December 2015 were analyzed.Realtime PCR was used to detect the expression levels of lncRNA FAM83 A-AS1 in 80 LUAD tissues and compare them with those in normal lung tissues adjacent to the cancer.Chi-square test was used to verify the relationship between lncRNA FAM83 A-AS1 level and clinicopathological features in LUAD patients.Overall survival(OS)was analyzed by Kaplan-Meier method;Cox regression model was applied to analyze the risk factors affecting the prognosis of LUAD.Results We found that lncRNA FAM83 A-AS1 was distinctly upregulated in LUAD tissues compared with the normal tissues(P<0.05).Higher level of lncRNA FAM83 A-AS1 was associated with tumor size,lymph nodes metastasis and TNM stage(P<0.05).Clinical investigation revealed that LUAD patients with high lncRNA FAM83 A-AS1 level showed a significantly shorter OS(P<0.05).COX regression survival analysis showed that lncRNA Fam83 A-AS1 gene expression was a risk factor for the prognosis of lung cancer,and its high expression indicated poor clinical prognosis in LUAD patients.Conclusion The present study suggested that the expression of lncRNA FAM83 A-AS1 in LUAD significantly increased,which is closely related to tumor size,lymph node metastasis,TNM stage and prognosis.
