摘 要:目的:观察参芪消痞汤对慢性萎缩性胃炎(CAG)癌前期病变大鼠胃黏膜表皮生长因子(EGF)、表皮生长因子受体(EGFR)表达水平与缺氧诱导因子(HIF-1α)、血管内皮生长因子(VEGF)及血清p53、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)的相关影响。方法:选择60只健康SD雄性大鼠喂养1周后,根据体重随机区组法将10只纳入正常组,50只纳入模型组。制备CAG癌前期病变大鼠模型,选取正常组中8只生长状况良好的大鼠纳入实验,选取与正常组存在显著差异的40只大鼠纳入实验,通过随机区组法将大鼠分为模型组、维酶素组、(低、中、高剂量)参芪消痞汤组,每组各8只。给予各组大鼠对应的药物灌胃治疗,正常组与模型组大鼠灌等体积的生理盐水,连续治疗3个月后检测各项指标表达水平。结果:与模型组对比,维酶素组及低、中、高剂量组大鼠胃黏膜组织病理损伤明显修复。各组大鼠胃黏膜EGF、EGFR表达水平均低于模型组(P<0.05)。模型组大鼠HIF-1α、VEGF表达水平高于其他各组(P<0.05),随着参芪消痞汤剂量的增加,大鼠HIF-1α、VEGF表达水平随之降低,且均低于模型组(P<0.05)。随着参芪消痞汤剂量的增加,大鼠p53、MDA逐渐下降,GSH-Px表达水平逐渐升高,与模型组差异统计学意义(P<0.05)。结论:参芪消痞汤可以降低胃黏膜EGF、EGFR、HIF-1α、VEGF、p53、MDA的表达水平,提高GSH-Px的表达水平,改善病变程度。Objective:To observe the expression levels of EGF and EGFR in gastric mucosa of rats with chronic atrophic gastritis(CAG),angiogenic factors HIF-1αand VEGF,related effects of serum p53,MDA and GSH-Px were investigated to explore their actual efficacy.Methods:After feeding 60 healthy SD male rats for 1 week,10 rats were included into the normal group and 50 into the model group according to the weight random zone group method.Utilize pylorus spring insert compound preparation of the CAG precancerous lesions in the rat model,selection of rats in normal group 8 rats in the experiment,known to grow only in good condition selection and normal group there were significant differences of 40 rats in the experiment,rats by randomized block divided into model group,the vatacoenayme group,(low,middle and high dose)ginseng and astragalus new soup group,each group of eight only.Rats in each group were given corresponding drug gavage treatment,and rats in the normal group and model group were given normal saline of the same volume,for 3 months.The expression levels of all indexes were measured.Results:Compared with the model group,the pathological damage of gastric mucosa in the vitexin group and the low,middle and high dose group was significantly repaired.The expression levels of EGF and EGFR in gastric mucosa of each group were lower than those of model group(P<0.05).The expression levels of HIF-1αand VEGF in model group were higher than those in other groups(P<0.05).With the increase of Shenqi Xiaopi decoction dose,the expression levels of HIF-1αand VEGF in rats decreased,which were lower than those in model group(P<0.05).With the increase of Shenqi Xiaopi decoction dosage,the expression levels of p53,MDA in rats gradually decreased,and were lower than those in model group(P<0.05).Conclusion:Shenqi Xiaopi decoction can reduce the expression levels of EGF,EGFR,HIF-1α,VEGF,p53 and MDA in gastric mucosa,increase the expression level of GSH-Px,and improve the degree of lesion.
