摘 要:目的观察腹腔注射天麻素对慢性偏头痛大鼠的治疗作用,并探讨其作用机制。方法取清洁级成年雄性SD大鼠24只,随机分为空白对照组、托吡酯组、模型组、天麻素组各6只。除空白对照组外,其他三组采用间歇性腹腔注射硝酸甘油(NTG)建立慢性偏头痛模型。注射NTG前30 min,模型组、托吡酯组、天麻素组分别给予腹腔注射生理盐水5 mL、托吡酯30 mg/kg、天麻素200 mg/kg,每日1次,共干预11 d;空白对照组不给予药物干预。干预第3、5、7、9、11天,用Vonfery电子机械痛测试仪、BW-Plantar 390足底热测痛仪测试给予NTG前后大鼠后爪的机械痛阈值与热痛阈值。干预第11天处死大鼠,取大鼠三叉神经节(TG)及三叉神经脊束尾侧核(TNC)。分别用RT-PCR法、Western blotting法检测TG、TNC中脑源性神经营养因子(BDNF)、降钙素基因相关肽(CGRP)mRNA及蛋白表达。结果与空白对照组比较,模型组不同时间点基础机械痛阈、基础热痛阈、急性机械痛阈、急性热痛阈低,行为学评分高,模型组BDNF、CGRP mRNA及蛋白相对表达量高(P均<0.05)。与模型组比较,托吡酯组、天麻素组不同时间点基础机械痛阈、基础热痛阈、急性机械痛阈、急性热痛阈高,行为学评分低,CGRP、BDNF mRNA及蛋白相对表达量低(P均<0.05)。天麻素组与托吡酯组以上指标比较差异无统计学意义(P均>0.05)。结论天麻素可能通过抑制初级感觉伤害感受器释放BDNF、CGRP,从而提高慢性偏头痛大鼠的痛阈,改善其症状。Objective To observe the therapeutic effect of intraperitoneal injection of gastrodin on chronic migraine rats and to explore its mechanism.Methods Twenty-four clean-grade adult SD male rats were randomly divided into the blank control group,topiramate group,model group,and gastrodin group,with 6 rats in each group.Except the blank control group,the rats in the other three groups were treated with intermittent intraperitoneal injection of nitroglycerin(NTG)to establish the models of chronic migraine.At 30 min before NTG injection,the rats in the model group,topiramate group and gastrodin group were intraperitoneally injected with normal saline 5 mL,30 mg/kg topiramate and 200 mg/kg gastrodin,once a day,for 11 days.On the 3rd,5th,7th,9th and 11th days of intervention,the mechanical pain threshold and thermal pain threshold of hind claws of rats before and after NTG were measured by Vonfery electromechanical pain tester and BW-Plantar390 plantar thermal pain tester.The rats were killed on the 11th day of intervention.The expression of brain-derived neurotrophic factor(BDNF),calcitonin gene-related peptide(CGRP)mRNA and protein in trigeminal ganglion(TG)and caudal nucleus of trigeminal nerve caudal nucleus were detected by RT-PCR and Western blotting,respectively.Results Compared with the blank control group,the basic mechanical pain threshold,basic thermal pain threshold,acute mechanical pain threshold,and acute thermal pain threshold were lower,the behavioral score was higher,and the relative expression of BDNF,CGRP mRNA and protein was higher in the model group at different time points(all P<0.05).Compared with the model group,the basic mechanical pain threshold,basic thermal pain threshold,acute mechanical pain threshold,and acute thermal pain threshold were higher,the behavioral score was lower,and the relative expression of CGRP,BDNF mRNA and protein was lower at different time points in the topiramate group and gastrodin group(all P<0.05).There was no significant difference in the above indexes between the ga
