Objective:To establish the patient-derived tumor xenografts(PDX)model of colorectal cancer(CRC),and explore the relationship among the anticancer effects, toxicity and dose of 5-fluorouracil(5-Fu).Methods:The fresh human CRC tissues were subcutaneously transplanted into immunodeficient mice to establish a PDX model, and the traceability of the test model was detected. The mice were treated with 5,10,20,40 and 60 mg/kg of 5-Fu,respectively,and the tumor volume, body mass and mortality were measured. Results:The PDX model was successfully established, and the traceability was 99.96%.The tumor inhibition rates of 5,10,20,40 and 60 mg/kg of 5-Fu were 33%,49%,68%,92%and 100%,respectively.Among them, the body weight of mice significantly lost in 40 mg/kg dose group, and the mortality of 60 mg/kg dose group was 100%.Conclusions:The efficacy and toxicity of 5-Fu are closely related to the doses, and the 10-20 mg/kg of 5-Fu is suitable for the efficacy evaluation of PDX model.
