摘 要:从广州徐闻农垦丰收农场土壤样品中分离到一株产羧甲基纤维素酶菌株,结合菌株形态特征和ITS基因序列同源性分析结果,表明该菌株为Lichtheimia sp.的未定种,其系统分类学关系与横梗霉菌Lichtheimia ramosa isolate K12(MN190291.1)最近,故命名为:横梗霉菌XWNR1(Lichtheimia ramosa XWNR1),该菌种保藏号为CCTCC NO:M2018230。采用刚果红染色法发现该菌粗酶液能够降解羧甲基纤维素钠产生透明圈,初步确定该菌具有产羧甲基纤维素酶的能力。通过优化该菌发酵培养条件,结果表明:该菌株以2.0×10^6接种浓度、1%接种量,28℃180 r/min恒温振荡培养60 h时,菌株分泌的羧甲基纤维素酶具有最好的活力。酶学性质初步研究结果表明,该羧甲基纤维素酶在最适pH 7.0、最适反应温度40℃条件下具有较好的pH稳定性和热稳定性。A carboxymethyl cellulase producing strain XWNR1 was isolated from the soil of Xuwen Nongken Harvest Farm in Guangzhou.The strain was identified as an undetermined species of Lichtheimia sp.based on the ITS sequence and the colony morphology.From the results of neighbor-joining tree,XWNR1 and Lichtheimia ramose isolate K12(MN190291.1)had the closest evolutional relationship.Therefore,it was named as Lichtheimia ramosa XWNR1,and the preservation number of the strain was CCTCC NO:M2018230.The Congo red staining method was used for screening strains with carboxymethyl cellulase activity,which indicated that the strain had the property of carboxymethyl cellulase.The fermentation medium and culture conditions of the strain was further optimized,and the results showed that when the strain was inoculated at 2.0×10^6 concentration and 1%inoculums size,and cultured at 28℃,180 r/min for 60 h,the carboxymethyl cellulase secreted by the strain had the highest crboxymethyl cellulase activity.The results of the preliminary study on its enzymatic properties revealed that the carboxymethyl cellulase had good pH stability and thermal stability at optimum pH 7.0 and reaction temperature of 40℃.
