摘　　要：目的探讨长链非编码RNA结肠癌相关转录物2(LncRNA-CCAT2)在结直肠癌(CRC)患者中的表达及其临床意义。方法收集2017年1月至2017年7月汉中市中心医院治疗的40例CRC患者的癌组织、癌旁组织、血清标本及40例无瘤患者的血清标本。运用实时荧光定量PCR(RT-qPCR)方法分别检测标本中LncRNA-CCAT2的表达量;采用受试者工作特征(ROC)曲线评价CCAT2对CRC的诊断价值;采用Kaplan-Meier分析CCAT2对CRC患者生存的影响。结果相较于癌旁正常组织标本,CCAT2在癌组织标本中表达明显上调(P<0.05);相较于无瘤患者的血清标本,CCAT2在CRC患者血清标本中表达上调,差异具有统计学意义(P<0.05)。当CCAT2联合CEA用于诊断CRC时,灵敏度、特异度、AUC分别为67.5%、87.5%、0.849。结论CCAT2可作为诊断及预测CRC预后的生物学标记物。Objective To investigate the expression and clinical significance of long non-coding RNA colon cancerrelated transcript-2(LncRNA-CCAT2)in patients with colorectal cancer(CRC).Methods From January 2017 to July 2017,cancer tissue,adjacent tissue,serum samples of 40 cases of CRC patients treated in Hanzhong central hospital and serum samples of 40 cases of non tumor patients were collected.The expression quantity of LncRNA-CCAT2 in the samples was detected by Real time quantitative PCR(RT-qPCR).The diagnostic value of CCAT2 for CRC was evaluated by receiver operating characteristic curve(ROC).The effect of CCAT2 on survival of CRC patients was analyzed by Kaplan-Meier.Results Compared with the normal tissue samples adjacent to cancer,the expression of CCAT2 in cancer tissue samples significantly up-regulated(P<0.05);compared with serum samples of non-tumor patients,the expression of CCAT2 in serum of CRC patients up-regulated,and the difference was statistically significant(P<0.05).When CCAT2 combined with CEA was used to diagnose CRC,the sensitivity,specificity and AUC were 67.5%,87.5%and 0.849,respectively.Conclusion CCAT2 can be a biomarker for the diagnosis and prognosis of CRC.
