摘 要:目的:探索不同剂量的次黄嘌呤和氧嗪酸钾(PO)灌胃构建高尿酸血症小鼠模型,筛选造模的最佳灌胃剂量。方法:选取50只雄性KM小鼠,随机分为空白组和4个模型组(A、B、C、D),模型组分别采用次黄嘌呤300mg/kg+氧嗪酸钾250mg/kg(A组)、次黄嘌呤300mg/kg+氧嗪酸钾750mg/kg(B组)、次黄嘌呤500mg/kg+氧嗪酸钾1000mg/kg(C组)、氧嗪酸钾1500mg/kg(D组)4种剂量连续灌胃7d,比较各组体重变化及血尿酸。结果:模型组和空白组比较,次黄嘌呤500mg/kg+氧嗪酸钾1000mg/kg灌胃组(C组)体重增长缓慢,造模后血清尿酸水平显著升高具有统计学意义(P<0.001),其余各模型组血清尿酸水平与空白组比较无统计学差异(P>0.05)。结论:次黄嘌呤500mg/kg联合氧嗪酸钾1000mg/kg连续灌胃给药7d,可建立高尿酸血症小鼠模型。Objective:To establish an efficient rat model of hyperuricemia with Hypoxanthine(HX)and Potassium Oxoxazine(PO).Methods:Fifty male KM rats were randomly divided into the blank group and four control groups(A,B,C,D),blank group intragastrical administration with Sodium carboxymethyl cellulose,control group A intragastrical administration with 300mg/kg HX and 250mg/kg PO,control group B intragastrical administration with 300mg/kg HX and 750mg/kg PO,control group C intragastrical administration with 500mg/kg HX and 1000mg/kg PO,control group D intragastrical administration with 1500mg/kg PO,all group Continuous intragastrical administration for 7 days.Compare the weight change of each group,blood was collected from the eye vein and the serum uric acid was detected.Results:Compared with the blank group,control group C reached high serum uric acid levels and has statistical significance(P<0.001).Conclusion:Intragastrical administration with 500mg/kg HX and 1000mg/kg PO for 7 days can establish a rat model of hyperuricemia.
