摘 要:目的:基于网络药理学初步筛选苗药定痛汤治疗偏头痛的作用靶点并探索其可能的分子机制。方法:通过TCMSP筛选苗药定痛汤潜在活性成分和作用靶点,利用TTD、GeneCards、OMIM数据库整理偏头痛疾病靶点,STRING数据库进行蛋白互作分析,Cytoscape_v3.8.0构建药物-疾病-靶点多维网络,再利用DAVID、ClueGO进行GO功能和KEGG进行基因富集分析。结果:运用TCMSP、TTD、GeneCards、OMIM,共筛选出苗药定痛汤129个潜在活性成分,2712个疾病靶点,共获得69个交集基因作为潜在治疗靶点;GO分析包含生物学过程582个条目、分子功能60个条目,细胞组分64个条目。KEGG通路富集筛选出73条关键通路,发现其作用机制可能与P13K/Akt、MAPK、NF-κB、SIRT1等多条信号通路相关。结论:苗药定痛汤具有多成分、多靶点、多途径的作用特点,对该方的药理机制研究及进一步的临床应用具有指导意义。Objective:To screen the target of Miao medicine(Dingtong decoction)in the treatment of migraine based on network pharmacology and explore its possible molecular mechanism.Methods:Screening potential active components and targets of Miao medicine(Dingtong decoction)by TCMSP,TTD、GeneCards、OMIM databases were used to sort out the targets of migraine,and STRING databases was used for protein interaction analysis,the multi-dimensional network of drug disease target was constructed by Cytoscape_v3.8.0,and then DAVID、ClueGO were used for go function and KEGG for gene enrichment analysis.Results:Using TCMSP,TTD,GeneCards,OMIM,129 potential active components and 2712 disease targets of Miao medicine(Dingtong decoction)were screened,and 69 intersection genes were obtained as potential therapeutic targets.GO analysis included 582 items of biological process,60 items of molecular function and 64 items of cell composition.73 key pathways were screened out by enrichment of KEGG pathway.The mechanism of Miao medicine(Dingtong decoction)may be related to multiple signaling pathways such as P13K/Akt,MAPK,NF-κB,SIRT1.Conclusion:Miao medicine(Dingtong decoction)has the characteristics of multi-component,multi-target and multi-channel action,which has guiding significance for the pharmacological mechanism research and further clinical application of the decoction.
