摘 要:目的观察根皮素对慢性束缚应激(chronic restraint stress,CRS)模型大鼠焦虑行为的影响及机制研究。方法 90只SD大鼠随机分为对照组(Con)、模型组(Mod)、舍曲林组(Ser)、根皮素低剂量组(Phl-L)、根皮素中剂量组(Phl-M)和根皮素高剂量组(Phl-H),每组15只。采用束缚器束缚大鼠(3 h/d,连续21 d)复制CRS大鼠模型。每日应激前30 min对各组大鼠进行灌胃给药,1次/d。Ser组灌胃盐酸舍曲林溶液15 mg/kg,Phl-L、-M、-H组分别灌胃根皮素溶液20 mg/kg、40 mg/kg、80 mg/kg,Con组和Mod组灌胃等量生理盐水。高架十字迷宫实验和旷场实验评价大鼠的焦虑行为,试剂盒检测大鼠海马组织中MDA含量和SOD、GSH-Px的活性,免疫蛋白印迹法检测大鼠海马组织Bcl-2和Bax的表达,免疫荧光组化观察大鼠海马组织Caspase-3的表达。结果与Con组比较,Mod组开臂进入次数百分比和开臂滞留时间百分比均明显减少(P<0.05),自主运动总距离明显缩短(P<0.05);大鼠海马组织中MDA含量以及Bax和Caspase-3蛋白表达明显升高(P<0.05),SOD和GSH-Px活性以及Bcl-2蛋白表达明显降低(P<0.05)。与Mod组比较,各给药组开臂进入次数百分比和开臂滞留时间百分比均明显增多(P<0.05),自主运动总距离明显延长(P<0.05);大鼠海马组织中MDA含量以及Bax和Caspase-3蛋白表达明显降低(P<0.05),SOD和GSH-Px活性以及Bcl-2蛋白表达明显升高(P<0.05)。结论根皮素可以改善CRS大鼠的焦虑行为,具有抗焦虑功能,其作用机制可能与抑制氧化应激、减少神经细胞凋亡有关。Objective To research the effects and mechanism of Phloretin on chronic restraint stress(CRS) model rats. Methods Ninety SD rats were randomly divided into control group(Con), model group(Mod), sertraline group(Ser), Phloretin-low dose group(Phl-L), Phloretin-middle dose group(Phl-M) and Phloretin-high dose group(Phl-H). Each group had 15 rats. Rats were placed in a restraint container(3 h daily, 21 d) to perform CRS modeling. The rats in each group were received intragastric administration 30 min before daily stress once a day. Rats in the Ser group were treated with sertraline hydrochloride solution at a dose of 15 mg/kg. Rats in Phl-L,-M,-H groups were treated with Phloretin solution at doses of 20 mg/kg, 40 mg/kg and 80 mg/kg, respectively. Rats in Con group and Mod group were treated with the equivalent normal saline. Elevated cross maze test and open field test were used to evaluate the anxious behaviors of the rats. The kit detected MDA content and the activity of SOD and GSH-Px in rat hippocampus. Expressions of Bcl-2 and Bax in hippocampus were determined by Western Blot. Immunofluorescence histochemistry was used to detect the expression of Caspase-3 in hippocampus. Results Compared with the Con group, percentages of entry number and retention time in open arm in Mod group were significantly decreased(P<0.05), total distance of autonomous movement significantly shortened(P<0.05), content of MDA and expressions of Caspase-3 and Bax in hippocampus significantly increased(P<0.05), while activity of SOD and GSH-Px and expression of Bcl-2 were significantly decreased(P<0.05). Compared with Mod group, percentages of entry number and retention time in open arm in each administration group rats were significantly increased(P<0.05), total distance of autonomous movement significantly lengthened(P<0.05), content of MDA and expressions of Caspase-3 and Bax in hippocampus significantly decreased(P<0.05), while activity of SOD and GSH-Px and expression of Bcl-2 were significantly increased(P<0.05).
