摘  要:目的:探究顺式阿曲库铵对人乳腺癌MCF-7细胞株生物学特性的影响。方法:体外培养人乳腺癌MCF-7细胞株,并分为对照组和顺式阿曲库铵5μmol/L组、10μmol/L组和20μmol/L组,分别使用对应剂量的顺式阿曲库铵预处理。克隆形成实验和EDU染色实验检测细胞增殖能力;流式细胞术和Hoechst 33342染色法检测细胞凋亡情况;Transwell法检测细胞侵袭能力;划痕实验检测细胞迁移能力;Western blot检测侵袭、迁移及凋亡相关蛋白E-cadherin、N-cadherin、Bax、Bcl-2、cytochrome C、PARP和cleaved PARP表达;免疫荧光法检测Viminten表达;将乳腺癌MCF-7细胞与人脐静脉内皮细胞共培养,检测节点数目,RT-PCR和Western blot检测VEGF表达。结果:与对照组相比,顺式阿曲库铵5μmol/L组细胞增殖、侵袭、凋亡及相关蛋白表达均无显著变化(P>0.05);顺式阿曲库铵10μmol/L组和20μmol/L组细胞增殖、侵袭能力显著减弱(P<0.05),细胞凋亡率显著升高(P<0.05);N-cadherin、VEGF、Bcl-2蛋白表达显著降低,E-cadherin、Bax、cytochrome C、PARP和cleaved PARP蛋白表达显著升高(P<0.05);VEGF mRNA表达及阳性斑点数显著降低(P<0.05),且呈剂量依赖性。结论:顺式阿曲库铵可显著抑制乳腺癌MCF-7细胞的增殖、侵袭及迁移并促进其凋亡,且在一定范围内呈剂量依赖性。Objective:To explore effects of cisatracurium on biological features of human breast cancer MCF-7 cell line.Methods:Human breast cancer MCF-7 cell lines were cultured in vitro and divided into control group,cisatracurium 5μmol/L group,10μmol/L group and 20μmol/L group,respectively,which were given corresponding dose of cisatracurium.Cell proliferation ability was detected by colony formation assay and EDU staining assay.Apoptosis was detected by flow cytometry and Hoechst 33342 staining.Cell invasion ability was detected by Transwell method.Cell migration ability was detected by scratch test.Expressions of invasion,migration and apoptosis-related proteins E-cadherin,N-cadherin,Bax,Bcl-2,cytochrome C,PARP and cleaved PARP were detected by Western blot.Expression of Viminten was detected by immunofluorescence.Breast cancer MCF-7 cells and human umbilical vein endothelial cells were co-cultured,and number of nodes were detected.Expression of VEGF was detected by RT-PCR and Western blot.Results:Compared with control group,there were no significant change in cell proliferation,invasion,apoptosis or expression of related protein in cisatracurium 5μmol/L group(P>0.05);cell proliferation and invasion ability in cisatracurium 10μmol/L group and 20μmol/L group were significantly decreased(P<0.05),while apoptosis rate was significantly increased(P<0.05).Expressions of N-cadherin,VEGF and Bcl-2 protein was significantly decreased while expression of E-cadherin,Bax,cytochrome C,PARP and cleaved PARP protein were significantly increased(P<0.05),expressions of VEGF mRNA and numbers of positive nodes were significantly decreased(P<0.05),and showing a dose-dependence.Conclusion:Cisatracurium can significantly inhibit proliferation,invasion and migration of breast cancer MCF-7 cells,and promote their apoptosis,and showing dose-dependence within certain dose range.
