摘 要:目的:探讨淫羊藿苷对胰腺癌细胞增殖和凋亡的作用及机制。方法:MTT法检测细胞活性确定给药浓度。对照组、淫羊藿苷12.5、25、50μmol/L组BxPC-3细胞用终浓度含有Icariin 0、12.5、25、50μmol/L培养基培养;顺铂10μmol/L组BxPC-3细胞用终浓度含有顺铂10μmol/L培养基培养;miR-9 inhibitor组和淫羊藿苷50μmol/L+miR-9 inhibitor组将转染有miR-9 inhibitor质粒的胰腺癌BxPC-3细胞用终浓度含有淫羊藿苷0或50μmol/L培养基培养。克隆形成实验检测细胞生长,流式细胞术检测细胞凋亡,蛋白印迹法检测Ki67、PCNA、cleaved caspase-3、Bax、Bcl-2蛋白相对表达水平,RT-qPCR检测miR-9 mRNA的表达。结果:选择淫羊藿苷12.5、25、50μmol/L做后续实验;淫羊藿苷减少胰腺癌BxPC-3细胞克隆细胞数目,下调增殖标记蛋白PCNA和Ki67表达,上调细胞凋亡率及凋亡标记蛋白Bax和Cleaved Caspase-3表达,下调Bcl-2表达,上调miR-9表达(P<0.01)。抑制miR-9表达逆转淫羊藿苷对胰腺癌细胞BxPC-3增殖和凋亡的作用。结论:淫羊藿苷抑制胰腺癌BxPC-3细胞增殖,并诱导BxPC-3细胞凋亡,这与miR-9上调有关。Objective:To investigate the effect and mechanism of icariin on the proliferation and apoptosis of pancreatic cancer cells.Methods:MTT assay was used to detect cell activity to determine the concentration.BxPC-3 cells in control group,Icariin 12.5μmol/L group,Icariin 25μmol/L group,Icariin 50μmol/L group contained Icariin 0,12.5,25,50μmol/L medium culture;BxPC-3 cells in Cisplatin 10μmol/L group were cultured in a medium containing Cisplatin 10μmol/L;pancreatic cancer BxPC-3 cells transfected with miR-9 inhibitor plasmids in miR-9 inhibitor group and Icariin 50μmol/L+miR-9 inhibitor group contain Icariin 0 or 50μmol/L medium culture.Cell growth was tested by clone formation experiments.Cell apoptosis was detected by flow cytometry.The relative expression levels of Ki67,PCNA,cleaved caspase-3,Bax,Bcl-2 proteins were detected by Western blot.The expression of miR-9 mRNA was detected by RT-qPCR.Results:Icariin 12.5,25 and 50μmol/L were selected for subsequent experiments.Icariin reduced the number of cloned cells of pancreatic cancer BxPC-3 cells,down-regulated the expression of proliferation marker proteins PCNA and Ki67,increased the rate of apoptosis and the expression of apoptosis marker proteins Bax and Cleaved Caspase-3,down-regulated the expression of Bcl-2,and up-regulated miR-9 expression(P<0.01).The inhibition of miR-9 expression reversed the effects of icariin on the proliferation and apoptosis of pancreatic cancer cells BxPC-3.Conclusion:Icariin inhibits the proliferation of pancreatic cancer BxPC-3 cells and induces apoptosis of BxPC-3 cells,which is related to the up-regulation of miR-9.
