摘　　要：目的旨在研究LncRNA-WTAPP1敲除对结肠癌细胞生长和运动能力以及裸鼠移植瘤的增生的影响。方法选择结肠癌SW480细胞进行研究。将shRNA1-WTAPP1,shRNA2-WTAPP1和 hRNA3-WTAPP1用Lipofectamine 2000转染到SW480细胞。选择转染,shRNA2-WTAPP1的细胞,建立对照组、shRNA-NC组和shKNA2-WTAPP1组,采用克隆形成试验,Transwall试验和Wound scratch试验分别检测结肠癌SW480细胞增殖、侵袭和迁移;采用qRT-PCR和Western印迹检测E-cadherin,N-cadherin和FN的相对表达量。将转染ARNA2-WTAPP1的SW480细胞注射裸鼠后腿的右侧腹从而建立原位异种移植小鼠模型。检测肿瘤的重量,并通过免疫组化检测E-cadherin和N-cadherin蛋白的表达量。结果LncRNA-W TAPP1 mRNA的表达量显著下调,尤其转染shRNA2-WTAPP1组。LncRNA-WTAPP1的低表达能抑制SW480细胞增殖、侵袭和迁移。在体内试验中,LncRNA-WTAPP1的低表达能抑制体内肿瘤的生长。结论LncRNA-WTAPP1敲除抑制结肠癌细胞生长和运动能力以及裸鼠移植瘤的增生。Objective This experiment aimed to study the effect of WTAPP1 knockout on the proliferation,invasion and migration of colorectal cancer cells and the orthotopic xenograft tumor in nude mice.Methods SW480 cells were used for this study.The shRNAl-WTAPP1,shRNA2-WTAPP1 and shRNA3-WTAPPl were transfected into SW480 cells by means of Lipofectamine 2000.A control group,an shRNA-NC group and an shRNA2-WTAPPl group were set up.The colony formation assay,Transwall assay and wound scratch assay were conducted to assess SW480 cells proliferation,invasion and migration,respectively.The relative expression of E-cadherin,N-cadherin and FN was detected by qRT-PCR and Western blotting.The SW480 cells were transfected with shRNA2-WTAPPl and were then injected into the right flank of the hind leg of nude mice to establish an orthotopic xenograft mouse model.Fhe weight of the tumor was determined,and the expression of E-cadherin and N-cadherin protein was detected by immunohistochemistry.Results shRNAl-WTAPP1,shRNA2-WTAPPl and shRNA3-WTAPPl were transfected into SW480 cells,and the mRNA expression level of WTAPP1 was significantly down-regulated,especially in the shRNA2-WTAPP1 group.The result displayed that low-expression of WTAPP1 inhibited SW480 cells proliferation,invasion and migration.In vivo experiments showed that low-expression of WTAPP1 inhibited tumor growth.Conclusion WTAPP1 knockout suppresses on the proliferation,invasion and migration of colorectal cancer cells and growth of orthotopic xenograft tumor in nude mice.
