摘 要:目的:探讨HLA-DRB1^(*)04位点下白细胞介素-34(IL-34)和集落刺激因子-1(CSF-1)表达水平对广西HCC家族聚集性的影响。方法:在广西HCC高发地区完成HLA-DRB1^(*)04位点检测的人群中收集HCC高发家族无癌成员51名作为高发家族成员组,直系血亲中未发生过任何肿瘤的家族成员49名作为无癌家族成员组。收集两组空腹外周血清样本,应用酶联免疫吸附试验(ELISA)技术检测研究对象外周血清IL-34和CSF-1的表达水平。分析HLA-DRB1^(*)04位点下IL-34和CSF-1表达水平对广西HCC家族聚集性的影响。结果:高发家族成员组IL-34表达水平显著高于无癌家族成员组(P<0.05);高发家族成员组CSF-1表达水平与无癌家族成员组比较,差异无统计学意义(P>0.05);HLA-DRB1^(*)04阳性组IL-34和CSF-1表达水平均显著高于HLA-DRB1^(*)04阴性组(P<0.05);HBsAg阳性组IL-34和CSF-1表达水平均显著低于HBsAg阴性组(P<0.05)。结论:IL-34高水平表达可能与广西HCC家族聚集性存在一定相关性。HLA-DRB1^(*)04等位基因导致广西HCC家族聚集性的发生可能是通过影响IL-34和CSF-1的高表达来实现。乙肝病毒(HBV)感染与IL-34和CSF-1的表达水平在广西HCC家族聚集中可能不具有协同作用。Objective:To investigate the effect of IL-34 and CSF-1 expressions at the HLA-DRB1^(*)04 site on the familial aggregation of HCC in Guangxi.Methods:51 cases of cancer-free members of high-risk families of HCC were collected from the population who had completed HLA-DRB1^(*)04 allele detection at the high incidence point of HCC in Guangxi as the high incidence family member group,and 49 lineal relatives without any tumor were selected as the cancer-free family member group.The peripheral serum samples of the two groups were collected,and the expression levels of IL-34 and CSF-1 in the peripheral serum of the subjects were detected by enzyme-linked immunosorbent assay(ELISA).In order to analyze the effect of IL-34 and CSF-1 expression level at HLA-DRB1^(*)04 locus on the aggregation of HCC family in Guangxi.Results:The expression level of IL-34 in high incidence family members group was significantly higher than that in cancer-free family member group(P<0.05),but there was no significant difference in CSF-1 expression between the two groups(P>0.05).The expression levels of IL-34 and CSF-1 in HLA-DRB1^(*)04 positive group were significantly higher than those in HLA-DRB1^(*)04 negative group(P<0.05).The expression levels of IL-34 and CSF-1 in HBsAg positive group were significantly lower than those in HBsAg negative group(P<0.05).Conclusion:The high expression level of IL-34 may be correlated with the familial aggregation of hepatocellular carcinoma in Guangxi.The occurrence of familial aggregation caused by the HLA-DRB1^(*)04 allele may be realized by influencing the high expression of IL-34 and CSF-1.HBV infection may not have a synergistic effect with the expression levels of IL-34 and CSF-1 in the Guangxi hepatocellular carcinoma family cluster.
