摘 要:目的比较肝癌细胞不同制悬液建立兔肝癌原位瘤模型,优选建模方法。方法取44只重量为2.6kg~3kg的新西兰大白兔,一方面将4只兔子后腿肌肉内注射VX2肿瘤细胞悬液,制作荷瘤种兔,无菌操作下分离并剪碎荷瘤种兔的肿瘤组织,收集细胞悬液。另一方面将40只兔子分为PBS缓冲液组(PBS组)、无血清DMEM溶液(EMEM组)、PBS和Matrigel混合组、无血清DMEM和Matrigel混合组,每组各10只兔子,经超声引导下观察其成瘤率。结果PBS和Matrigel混合组成瘤率为70%;无血清DMEM和Matrigel混合组的成瘤率为90%;PBS缓冲液组的成瘤率为30%;无血清DMEM溶液的成瘤率为50%,差异有统计学意义(p<0.05)。结论采用DMEM和Matrigel混合制作兔VX2肝癌细胞稀释制悬后经超声引导下可以最有效建立兔肝癌原位瘤模型。Objective:To compare of rabbits VX2 live tumor model with different cell suspensions for optimum modeling methods.Methods:Totally 44 New Zealand white rabbits weighing 2.6 kg~3 kg were selected as the objects of study.On one hand 4 rabbits’were performed intramuscular injection of VX2 cell suspensions as samples,and the cell suspensions were extracted.On other hand,40 rabbits were divided into the group of PBS(n=10),the group of EMEM(n=10),the group of PBS and Matrigel(n=10),the group of EMEM and Matrigel(n=10),and the successful rate to establish the rabbit VX2 liver tumor model was observed by ultrasound.Results:The successful rate was 70%in the group of PBS and Matrigel,90%in the group of DMEM and Matrigel,30%in the group of PBS,and 50%in the group of DMEM,with statistical significance(p<0.05).Conclusions:The highest successful rate to establish the rabbit VX2 liver tumor model was used cell suspensions of DMEM and Matrigel.
