摘　　要：目的探讨蛋白磷酸酶2A调节亚基B''α(protein phosphatase 2 regulatory subunit B″alpha,PPP2R3A)在乙肝相关性肝癌中的表达及临床意义。方法利用免疫组织化学方法检测PPP2R3A蛋白在70例乙肝相关性肝癌和40例良性肝病组织中的表达,采用酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)法检测PPP2R3A在肝癌患者外周血中的蛋白水平表达,运用统计学方法分析PPP2R3A蛋白水平的表达与乙肝相关性肝癌临床病理学指标的关系。结果PPP2R3A蛋白在肝癌组织中的表达主要定位于细胞质,其在肝癌组织中的表达明显高于癌周组织(P<0.001),并且PPP2R3A蛋白在肝癌组织中的表达明显高于良性肝病(P<0.001)。PPP2R3A蛋白的表达强度与肝癌患者的AFP值(r=0.256,P=0.032)具有统计学差异。ELISA结果也显示PPP2R3A蛋白在肝癌患者外周血中的表达量明显高于其在良性肝病中的表达(P=0.033),与免疫组织化学结果一致。结论PPP2R3A蛋白在乙肝相关性肝癌组织及血清中呈高表达,且与肝癌的临床病理特征有相关性,可能与肝癌的发生发展有关。Objective To investigate the expression and clinical significance of protein phosphatase 2 regulatory subunit B'α(protein phosphatase 2 regulatory subunit B"alpha,PPP2R3A)in hepatitis B virus(HBV)related hepatocellular carcinoma(HCC).Methods The expression of PPP2R3A protein in 70 cases of HBV related HCC and 40 cases of benign liver disease was detected by immunohistochemical method.The expression level of PPP2R3A in peripheral blood of patients with HCC was detected by enzyme linked immunosorbent assay(ELISA)method.The correlation between the expression of PPP2R3A protein and clinicopathological indexes of HBV related HCC was analyzed by statistical method.Results The expression of PPP2R3A protein in HCC tissues was mainly located in the cytoplasm,and the expression of PPP2R3A protein in HCC tissues was significantly higher than that in peri-cancerous tissues(P<0.001),and the expression of PPP2R3A protein in HCC tissues was significantly higher than the liver tissue of benign liver diseases(P<0.001).The expression intensity of PPP2R3A protein was statistically different from the AFP value of HCC patients(r=0.256,P=0.032).The results of ELISA also showed that the expression of PPP2R3A protein in peripheral blood of patients with HCC was significantly higher than that in benign liver diseases (P = 0.033), which was consistent with the results of immunohistochemistry. Conclusion PPP2R3A protein ishighly expressed in the tissue and serum of HBV related HCC, and is related to the clinicopathological features ofHCC, which may be related to the occurrence and development of HCC.
