摘  要:目的探讨结直肠癌组织中miR-99a、miR-100的表达水平与肿瘤转移的关系。方法收集2014年1月至2019年1月台州市第一人民医院收治的84例行手术治疗的结直肠癌患者的肿瘤组织、癌旁组织和正常组织标本,采用实时荧光定量PCR法检测各组织中miR-99a、miR-100的表达水平,收集患者的临床病理参数及随访情况,统计分析miR-99a、miR-100的表达水平与患者临床病理学特征及总体生存率的关系。结果miR-99a在结直肠癌组织、癌旁组织、正常组织中的相对表达量分别为3.15±1.52、5.98±1.73、8.63±1.98,miR-100的相对表达量分别为2.86±1.05、5.64±1.47、8.52±1.56。结直肠癌组织中miR-99a、miR-100的相对表达量显著低于癌旁组织及正常组织,差异均有统计学意义(P均<0.05)。84例患者中有23例出现肿瘤复发或转移。Log-rank生存分析发现,miR-99a低表达组的总体生存率显著低于miR-99a高表达组,miR-100低表达组的总体生存率显著低于miR-100高表达组,差异均具有统计学意义(P均<0.05)。Cox生存回归分析发现,Duke′s分期、miR-99a、miR-100是影响患者复发或肿瘤转移的独立危险因素(P<0.05)。结论结直肠癌患者组织中miR-99a、miR-100的表达被显著抑制,其低表达对患者的临床病理学特征有显著的不良影响。miR-99a和miR-100具有潜在的抑癌作用。Objective An attempt is made in this paper to investigate the relationship between the expressions of miR-99a,miR-100 and tumor metastasis in colorectal cancer.Methods The tumor tissues,the adjacent tissues,and the normal tissues of 84 patients with colorectal cancer who were admitted to Taizhou First People′s Hospital from January 2014 to January 2019 for surgical treatment were collected.The expressions of miR-99a and miR-100 in those tissues were detected by real-time fluorescent quantitative PCR.The clinicopathological parameters and follow-up date were collected to statistically analyze the relationship between the expressions of miR-99a,miR-100 and the clinicopathological features and overall survival rate of the patients.Results The relative expressions of miR-99a and miR-100 were(3.15±1.52,5.98±1.73,8.63±1.98)and(2.86±1.05,5.64±1.47,8.52±1.56)respectively in colorectal cancer tissues,adjacent tissues,and normal tissues.The relative expressions of miR-99a and miR-100 in colorectal cancer tissues were significantly lower than those in adjacent tissues and normal tissues(P<0.05).Twenty-three out of 84 patients had tumor recurrence or metastasis.Log rank survival analysis showed that the overall survival rate of the low miR-99a expression group and the low miR-100 expression group were significantly lower than those of the high miR-99a expression group and the high miR-100 expression group(P<0.05).Cox regression analysis showed that Duke′s stage,expressions of miR-99a and miR-100 were independent risk factors for tumor recurrence or metastasis(P<0.05).Conclusions The expressions of miR-99a and miR-100 in colorectal cancer tissues are significantly inhibited.Low expressions of miR-99a and miR-100 have significant adverse effects on clinicopathological characteristics of colorectal cancer patients,which have a potential anti-cancer effect.
