摘  要:目的探讨人组织激肽释放酶家族(KLK)在结直肠癌中的表达及其对预后的影响。方法挖掘ONCOMINE数据库,对各种类型恶性肿瘤中人KLK基因的表达进行分析。挖掘TCGA数据库,分析人KLK基因在结直肠癌组织和正常结直肠组织中的表达情况,进一步挖掘TCGA数据库以及GSE39582系列数据,分析人KLK基因表达与结直肠癌预后的关系。通过定量逆转录聚合酶链反应分析临床样本中人KLK基因在结直肠癌组织和正常结直肠组织中的表达水平。通过挖掘STRING数据库,对KLK基因相关的蛋白质-蛋白质作用网络进行分析。结果 ONCOMINE数据库中,KLK6、KLK8、KLK10和KLK11基因在结直肠癌组织中具有高水平的表达。GSE39582系列数据中,结直肠癌组织中人KLK6、KLK7、KLK8、KLK10、KLK11和KLK12基因的表达水平均明显高于正常结直肠组织,差异均有统计学意义(P﹤0.01)。KLK6、KLK8、KLK10和KLK12基因低表达患者的预后分别优于上述基因高表达的患者,差异均有统计学意义(P﹤0.05)。临床样本中,结直肠癌组织中KLK1基因的表达水平低于正常结直肠组织,KLK6、KLK7、KLK8、KLK10和KLK11基因的表达水平均高于正常结直肠组织,差异均有统计学意义(P﹤0.05)。通过蛋白质-蛋白质作用网络分析发现,α-突触核蛋白(SNCA)、YY1关联因子2(YAF2)和α2-巨球蛋白样蛋白1(A2ML1)等10个蛋白与KLK6、KLK8和KLK10具有相互作用。结论 KLK6、KLK8和KLK10基因在结直肠癌组织中高表达,且与结直肠癌患者的预后有关。Objective To investigate the expression of human kallikrein-related peptidase(KLK)in colorectal cancer and its influence on prognosis.Method The ONCOMINE database was mined to analyze the expression of the human KLK gene m various types of malignant tumors.Further,TCGA database was used to analyze the expression of the human KLK gene m colorectal cancer tissues and normal colorectal tissues.The TCGA database and GSE39582 series were taken together to explore the relationship between human KLK gene expression and the prognosis of colorectal cancer.Quantitative reverse transcription-polymerase chain reaction(qRT-PCR)was used to venfy and analyze the expression level of the human KLK gene in colorectal cancer tissues and normal colorectal tissues in clinical samples.By exploring the STRING database,the protein-protein interaction network related to KLK gene was also made.Result According to the ONCOMINE database,KLK6,KLK8,KLK10 and KLK11 genes have hight expression levels in colorectal cancer tissues.Based on the GSE39582 series,the expression levels of human KLK6,KLK7,KLK8,KLK10,KLK11 and KLK12 genes in colorectal cancer tissues were significantly higher than those in normal colorectal tissues,and the differences were statistically significant(P<0.01).The prognosis of patients with low expression of KLK6,KLK8,KLK10 and KLK12 genes was better than that of patients with high expression of the forgoing genes,and the differences were statistically significant(P<0.05).According to the verified cohort,the expression level of the KLK1 gene in colorectal cancer tissue was lower than that in normal colorectal tissue,the expression level of KLK6,KLK7,KLK8,KLK10 and KLK11 genes was significantly higher than that in normal colorectal tissue,the differences were statistically significant(P<0.05).According to the protein-protein interaction network analysis,it is evident that ten proteins such as alpha-synuclein(SNCA),YY1 associated factor 2(YAF2),and alpha-2-macroglobulin like protein 1(A2 ML1)interact with KLK6,KLK8,and KLK10.
