卵巢癌患者miR-497、miR-125a-5p的表达及与其临床特征和生存率的关系

摘　　要：目的:分析卵巢癌患者癌组织中微小RNA-497(miR-497)和微小RNA-125a-5p(miR-125a-5p)的表达情况及其临床意义。方法:选取2018-06—2019-12在本院手术治疗的96例卵巢癌患者作为研究对象,将手术切除的卵巢癌组织作为试验组,癌旁(>2cm)正常组织为对照组。采用实时荧光定量PCR(qRT-PCR)检测组织中miR-497和miR-125a-5p的表达情况;比较不同临床病理特征卵巢癌患者miR-497和miR-125a-5p表达水平的差异;分析卵巢癌患者三年生存率与miR-497和miR-125a-5p水平的关系;分析影响卵巢癌患者生存率的危险因素。结果:试验组miR-497和miR-125a-5p的表达量均明显低于对照组(P<0.01);与Ⅰ-Ⅱ期卵巢癌患者相比,Ⅲ-Ⅳ期患者miR-497、miR-125a-5p表达水平均较低(P<0.01),与无淋巴结转移患者相比,淋巴结转移者miR-497、miR-125a-5p表达水平均较低(P<0.05);miR-497、miR-125a-5p低表达组生存率均明显低于高表达组(P<0.05);COX回归分析显示miR-497低表达及miR-125a-5p低表达是影响卵巢癌患者生存率的独立危险因素。结论:卵巢癌患者癌组织中miR-497和miR-125a-5p表达下调,其表达水平与卵巢癌患者三年生存率密切相关。Objective:To study the expressions of microRNA-497(miR-497)and microRNA-125a-5p(miR-125a-5p)in ovarian cancer tissues and their clinical significances.Method:96 patients who underwent surgery in our hospital from June 2018 to December 2019 were selected as the study subjects.The ovarian cancer tissues resected by surgery were taken as the experimental group,and the normal tissues adjacent to the cancer(>2cm)were taken as the control group.Real-time fluorescence quantitative PCR(qRT-PCR)was used to detect the expressions of miR-497 and miR-125a-5p,the relationships between miR-497,miR-125a-5p and clinicopathological features of ovarian cancer were analyzed,the relationships between the prognosis of ovarian cancer patients and the expressions of miR-497 and miR-125a-5p were analyzed,the factors influencing the prognosis of ovarian cancer patients were analyzed by COX regression analysis.Results:The expressions of miR-497 and miR-125a-5p in the experimental group were significantly lower than those in the control group(P<0.01).The expression levels of miR-497 and miR-125a-5p in patients with stageⅢ-Ⅳwere lower than those in patients with stageⅠ-Ⅱ(P<0.05).The expression levels of miR-497 and miR-125a-5p in patients with lymph node metastasis were lower than those in patients without lymph node metastasis(P<0.05).The survival rates of miR-497 and miR-125a-5p low expression groups were significantly lower than those of high expression group(P<0.05).COX regression analysis showed that low expression of miR-497 and miR-125a-5p were independent risk factors affecting the prognosis of patients with ovarian cancer.Conclusion:The expressions of miR-497 and miR-125a-5p in ovarian cancer tissue are down-regulated,the expression level of miR-497 and miR-125a-5p are closely related to the prognosis of ovarian cancer patients and have clinical value.
