摘  要:金黄色葡萄球菌和链球菌是奶牛乳腺炎的重要致病菌,为了研究预防金黄色葡萄球菌和链球菌感染的奶牛乳腺炎新型疫苗,以金黄色葡萄球菌FnBPA的1个CD4^(+)T细胞表位和两个B细胞表位构建了FTB1B2表位疫苗,以停乳链球菌GapC的1个CD4^(+)T细胞表位和两个B细胞表位构建了GTB1B2表位疫苗,再以GSGSGS作为Linker,串联FTB1B2与GTB1B2构建了表位疫苗FG;串联了FnBPA的CD4^(+)T细胞表位FT和GapC的CD4^(+)T细胞表位GT构建了表位疫苗FGT。通过免疫接种BALB/c小鼠评价疫苗的免疫原性。结果显示,FT、GT和FGT仅能诱导细胞免疫应答,并不能显著抑制金黄色葡萄球菌或停乳链球菌在肝、脾、肺、肾各器官的定殖数;FG诱导细胞和体液免疫应答能力显著高于GT、FGT和GST,显著低于FTB1B2和GTB1B2;FG免疫小鼠各器官细菌定殖数显著低于FT、GT、FGT和GST,显著高于FTB1B2和GTB1B2,但能够同时抑制金黄色葡萄球菌和停乳链球菌在各器官的定殖。结果表明,研究构建的FG表位疫苗能够诱导良好的免疫应答,并能同时抑制攻毒小鼠器官中金黄色葡萄球菌和停乳链球菌的定殖。Staphylococcus aureus and streptococcus were important pathogens of dairy cow mastitis.To study epitope-vaccines against dairy mastitis caused by Staphylococcus aureus and streptococcus,multi-epitope vaccines containing CD4^(+)T cell epitopes and B cell epitopes from FnBPA and GapC were constructed for prevention of S.aureus and/or streptococcus infections.Based on a CD4^(+)T cell epitope and two B cell epitopes from FnBPA,an epitope-vaccine FTB1B2 was constructed,and based on a CD4^(+)T cell epitope and two B cell epitopes from GapC,an epitope-vaccine GTB1B2 was constructed.Using GSGSGS as the Linker,FTB1B2 and GTB1B2 were connected in tandem to form a novel multiepitope-vaccine FG,and a CD4^(+)T cell epitope FT from FnBPA and another CD4^(+)T cell epitope GT from GapC were connected in tandem to form an epitope-vaccine FGT.BALB/c mice were immunized with the epitope-vaccines for the evaluation of immune efficacy.The results showed that FT,GT and FGT could induce only cellular immune responses and could not significantly inhibit colonization of S.aureus and S.dysgalactiae in various organs of liver,spleen,lung and kidney.FG induced significantly higher cellular and humoral immune responses than GT,FGT and GST,but significantly lower than FTB1B2 and GTB1B2;the numbers of bacterial colonization in the organs of the mice immunized with FG were lower than those of the mice immunized with FT,GT and FGT,but higher than those of the mice immunized with FTB1B2 and GTB1B2.The above results indicated that the constructed FG could induce an ideal immune response and inhibit colonization of S.aureus and S.dysgalactiae in the organs of challenged mice.
