摘　　要：目的研究喹硫平(quetiapine,QTP)对精神分裂症模型小鼠脑内星形胶质细胞和小胶质细胞的作用,探讨喹硫平抗精神病的受体外药理作用机制。方法48只雄性C57BL/6小鼠分为QTP+铜腙(cuprizone,CPZ)组、生理盐水(normal saline,NS)+CPZ组、QTP组、NS组等4组,每组12只。通过旷场实验评测小鼠的自发活动,用免疫荧光标记的anti-胶质纤维样酸性蛋白(glia fibrillary acidic protein,GFAP)抗体和anti-钙离子连接蛋白分子-1(calcium bingding adaptor molecule-1,Iba-1)抗体分别检测脑内胼胝体、皮质和海马CA1区星形胶质细胞和小胶质细胞的数量。结果旷场实验中QTP+CPZ组小鼠总路程比NS+CPZ组减少(P<0.01);NS+CPZ组与NS组相比总路程增多(P<0.05)。QTP+CPZ组和NS+CPZ组比较,脑内GFAP(P<0.05)和Iba-1(P<0.01)阳性细胞均减少;NS+CPZ组与NS组相比,脑内GFAP(P<0.01)和Iba-1(P<0.01)阳性细胞增多。结论QTP能够减轻CPZ诱导的精神分裂症小鼠脑内胶质细胞增生。Objective This study aimed at examining effects of quetiapine(QTP),an atypical antipsychotic,on the behaviors and brain gliocyte proliferation in cuprizone(CPZ)-induced mice.Methods Forty eight male young(6-weeks old)C57BL/6 mice were randomly assigned to four groups:QTP+CPZ,NS+CPZ,QTP and NS groups.The mice first had the diet with or without CPZ(0.2%,w/w)for 7 days and then receiving injection of QTP(10 mg/kg/d)in the saline or sterilized saline(1 mL/100 g)for additional two weeks.The mice were subjected to the open-field tests one day after the last injection.After sacrifice,their brains were processed for morphological analyses(immunofluorescence staining).Results CPZ-fed mice traveled a longest total distance(TD)among the four groups(P<0.05).Statistic revealed significant differences between QTP+CPZ and NS+CPZ groups(P<0.01).Mice in NS+CPZ group showed significant increases in the number of GFAP(P<0.01)and Iba-1(P<0.01)positive cells compared to NS group,which was attenuated by QTP.Conclusions Quetiapine can attenuate the gliocyte proliferation by CPZ-exposure.
