慢性铅中毒小鼠抑郁样症状与脑额叶中5-HT含量的关系研究

摘　　要：目的探讨小鼠慢性铅中毒是否通过影响脑额叶中5-羟色胺(5-HT)含量来诱发抑郁样症状。方法5周龄健康雄性小鼠分为对照组和实验组适应性喂养1周后,实验组的3组实验亚组分别腹腔注射醋酸铅5、7、12mg/kg,对照组注射等量的生理盐水,连续2周,建立慢性铅中毒小鼠模型。用蔗糖水偏好实验、旷场实验来评估抑郁样症状。采用ELISA试剂盒法、原子吸收光谱法分别测定额叶组织5-HT含量、全血标本铅含量。结果偏好测试、旷场测试结果显示,实验组小鼠出现抑郁样症状,偏好测试结果各组比较,差异均有统计学意义(P<0.05);低、中、高实验组血铅含量依次升高,额叶5-HT含量依次减低,差异均有统计学意义(P<0.05)。结论慢性铅中毒小鼠存在抑郁样症状,且这种抑郁样症状可能是由5-HT的表达失衡引起的。Objective To investigate whether chronic lead poisoning can induce depressive-like symptoms by affecting serotonin(5-HT)level in the frontal lobe of the brain of mouse.Methods Five-weekold healthy male mice were divided into control group and experimental group in one week with adaptive feeding.Three experimental subgroups were injected intraperitoneally with lead acetate 5,7,12mg/kg,and the control group was injected with the same amount of normal saline.After two consecutive weeks,the mouse model of chronic lead poisoning were established.Depression-like symptoms were evaluated by sucrose water preference test and open field test.The contents of 5-HT in frontal lobe tissue and lead in whole blood were determined by Elisa Kit and atomic absorption spectroscopy respectively.Results The results of preference test and open field test showed that the mice in the experimental group had depressive-like symptoms,and the levels of blood lead in the low,middle and high test groups were significantly increased(P<0.05)The content of 5-HT in frontal lobe of low,middle and high experimental group decreased in turn,the difference was significant(P<0.05).Conclusion Chronic lead poisoning mice have depression-like symptoms which may be caused by an imbalance in the expression of 5-HT.
