摘  要:目的:通过加权基因共表达网络分析(WGCNA)方法筛选促进乳腺癌循环肿瘤细胞转移的基因,为乳腺癌转移的早期预警提供循环肿瘤细胞标志物,也为乳腺癌发展动态监测提供新策略。方法:利用WGCNA筛选出数据集GSE9893中与乳腺癌转移相关的基因,将所选基因与在循环肿瘤细胞数据库中高表达的基因取交集作为候选基因,最后在肿瘤基因组计划(TCGA)数据库中验证候选基因的表达水平。结果:1024个基因在伴转移的乳腺癌中高表达,WGCNA从中得到与乳腺癌转移相关的67个基因,其中ACTB和DDX5基因在乳腺癌循环肿瘤细胞中呈现出高表达的状态,并且DDX5在TCGA伴转移的乳腺癌样本中也高表达。结论:DDX5可作为乳腺癌发展动态监测的循环肿瘤细胞标志物,并可能为乳腺癌治疗提供新的靶点。Objective:To find genes that promote the metastasis of breast cancer circulating tumor cells(CTCs)through the weighted gene co-expression network analysis(WGCNA)method,provide circulating tumor cell markers for early warning of breast cancer metastasis,and also provide new strategies for breast cancer development dynamic monitoring.Methods:Genes related to breast cancer metastasis in the data set GSE9893 were selected by WGCNA.And the CTCs database was used to further find out the genes that are highly expressed in CTCs of breast cancer.Finally,data from the Cancer Genome Atlas(TCGA)breast cancer were used to verify the expression of selected genes.Results:1024 genes were highly expressed in breast cancer samples with metastasis,among which 67 genes were found to be related to breast cancer metastasis through WGCNA.Among the 67 genes,ACTB and DDX5 were also highly expressed in CTCs of breast cancer.And DDX5 is also highly expressed in TCGA breast cancer samples with metastasis.Conclusion:DDX5 can be used as a circulating tumor cell marker for dynamic monitoring of breast cancer development,and may provide a new target for breast cancer treatment.
