摘  要:为了驯化活性污泥中产聚羟基脂肪酸酯(PHA)的混合菌群并解析细菌群落结构变化,该研究采用好氧瞬时补料(ADF)工艺以乙酸钠为碳源驯化活性污泥合成PHA,并运用高通量测序分析细菌群落结构变化。结果表明,pH 7.0、乙酸钠4 g/L驯化出的混合菌群PHA合成能力最强;积累的PHA为聚羟基丁酸酯(PHB),含量达到14.72%,浓度达到1.92 g/L。高通量测序分析表明驯化后的污泥细菌群落多样性远低于驯化前的污泥,说明驯化过程中一部分细菌被淘汰。不同分类水平上的细菌群落分析表明驯化前后优势细菌类群发生了较大的变化。驯化后的污泥细菌群落中优势菌属包括噬氢菌属(Hydrogenophaga)、黄杆菌属(Flavobacterium)、短波单胞菌属(Brevundimonas)和陶厄氏菌属(Thauera),均为已报道的PHA积累菌属。A bench-scale experiment was conducted aiming at enrichment of the polyhydroxyalkanoate(PHA)-accumulating microorganisms in activated sludge,and meantime for probing into the variation of bacterial community structure.Experiment work done involved the PHA-accumulating microorganisms that were acclimatized and enriched in the activated sludge by the aerobic dynamic feeding(ADF)technology with sodium acetate as a sole carbon source.Besides,the bacterial community structure was studied by the high-throughput sequencing as well.The experimental findings showed that when culture solution being pH 7.0 and sodium acetate added being 4 g/L,the mixed microorganisms cultured had the highest level of PHA;the accumulated PHA form belonged to polyhydroxybutyrate(PHB)with the PHB content being up to 14.72%,and the PHB concentration up to 1.92 g/L.High-throughput sequencing analysis revealed that the bacterial diversity of enriched sludge was lower than that of original sludge,indicating that some bacterial taxa had been eliminated during enrichment,and the dominant bacterial population changed considerably after enrichment.The dominant genera in the enriched sludge constituted Hydrogenophaga,Flavobacterium,Brevundimonas and Thauera.All of them belonged to PHA-accumulating bacterial genera,which had been reported in related publications.
