涎腺多形性腺瘤细胞中RECK和MMP-9的表达及与肿瘤复发的相关性分析

摘　　要：目的:分析回复引导半胱氨酸丰富蛋白Kazal基元(reversion inducing cysteine rich protein with Kazal motifs,RECK)和基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)的表达及其与涎腺多形性腺瘤(pleomorphic adenoma,PA)肿瘤复发的相关性。方法:采用荧光实时定量PCR和细胞爬片方法检测15例涎腺初发型PA与19例涎腺复发型PA中RECK和MMP-9的转录与蛋白表达水平。结果:PCR结果显示:复发型PA细胞中RECK mRNA表达均显著低于初发型,而MMP-9 mRNA在复发型PA细胞中均显著高于初发型PA,结果均具有统计学差异(P<0.05)。初发型与复发型PA细胞中RECK/MMP-9比值分别为15.89、0.33。RECK/MMP-9比值从15.89下降至0.33。细胞爬片发现,复发型PA细胞中RECK表达低于初发型,而MMP-9在初发型PA细胞中阳性表达低于复发型,其结果均具有统计学差异(P<0.05)。初发型与复发型PA细胞中RECK/MMP-9比值分别为1.76、0.60,RECK/MMP-9比值从1.76下降至0.60。结论:RECK和MMP-9在涎腺初发型与复发型PA细胞中的表达差异显著,可能与涎腺PA的复发相关。Objective:To investigate the correlation between the expression of RECK and MMP-9 and the recurrence of salivary gland pleomorphic adenoma tumors.Methods:Transcription and protein expression levels of RECK and MMP-9 in 15 cases of salivary glandular pleomorphic adenomas and 19 cases of recurrent pleomorphic adenomas were detected by fluorescence real-time quantitative PCR and cell climbing.Results:The PCR results showed that the expression of RECK mRNA in recurrent PA cells was significantly lower than that in the initial type,and the mRNA of MMP-9 in recurrent PA cells was significantly higher than that in the initial type.The results were statistically significant(P<0.05).The ratios of RECK/MMP-9 in priming and recurrent PA cells were 15.89 and 0.33.The RECK/MMP-9 ratio decreased from 15.89 to 0.33.Cell crawling showed that the expression of RECK in the recurrent PA cells was lower than that in the initial type,while the positive expression of MMP-9 in the initial PA cells was lower than that in the recurrent type.The results were statistically significant(P<0.05).The ratios of RECK/MMP-9 in primal and recurrent PA cells were 1.76 and 0.60.The RECK/MMP-9 ratio decreased from 1.76 to 0.60.Conclusion:The expression of RECK and MMP-9 are significantly different in salivary glandular and recurrent pleomorphic adenomas.Which may be related to the recurrence of salivary glands pleomorphic adenomas.
