摘  要:目的:分析甲状腺乳头状癌经典型(CPTC)与滤泡型(FVPTC)基因表达谱差异,并探究相关差异表达基因(DEGs)对淋巴结转移及预后的影响。方法:从基因表达综合数据库(GEO)获得含两种亚型甲状腺乳头状癌(PTC)和正常组织的表达谱数据,应用R语言进行差异基因筛选及差异显著DEGs热图绘制,使用Cytoscape软件进行生物学功能富集、信号通路和核心基因筛选,利用UALCAN在线网站分析核心基因对PTC组织病理、转移及预后的影响。结果:共得到81个DEGs在两种亚型中共同表达,各有261个和78个DEGs仅存在于CPTC和FVPTC中。CPTC主要涉及酪氨酸代谢和p53信号通路,FVPTC主要富集于酪氨酸代谢和PPAR信号通路。细胞外基质(ECM)相关核心基因与PTC淋巴结转移及预后显著相关。结论:FVPTC和CPTC涉及的DEGs及通路既有相同点也存在一定差异,ECM基因可能是鉴别PTC亚型、预测淋巴结转移和预后的潜在标记物。Objective:To analyze the difference of gene expression profiles in classical variant(CPTC)and follicular variant(FVPTC)papillary thyroid carcinoma(PTC)and explore the effects of related differentially expressed genes(DEGs)on lymph node metastasis and prognosis.Methods:The gene expression profile datum of tumor and normal tissues of the two subtypes were obtained from the comprehensive gene expression database(GEO).R language was used to screen DEGs and draw heatmap according to significantly differented DEGs.Cytoscape software was applyed to biological function enrichment,signal pathway and key genes screening.UALCAN online website was used to analyze the function of core genes in PTC pathology,metastasis and prognosis.Results:A total of 81 DEGs were differently expressed both in the two subtypes,and 261 and 78 DEGs were only expressed in CPTC and FVPTC respectively.Tyrosine metabolism and p53 signal pathway functioned in CPTC,while tyrosine metabolism and PPAR signal pathway in FVPTC.Core extracellular matrix(ECM)genes were remarkably associated with lymph node metastasis and prognosis of PTC.Conclusion:There are both similarities and differences in DEGs and pathways between FVPTC and CPTC.ECM genes may be potential markers for distinguishing PTC subtypes and predicting lymph node metastasis and prognosis.
