摘  要:目的:探讨脐血来源的树突状细胞(DC)增强自然杀伤(NK)细胞和自然杀伤性T(NKT)细胞对白血病细胞杀伤作用的影响机制。方法:提取脐带血单个核细胞(PBMC),体外诱导培养DC、NK和NKT淋巴细胞。以人髓性白血病K-562细胞作为靶细胞,检测NK、NKT、NK+DC和NKT+DC对靶细胞的杀伤效应。检测共培养上清液中IFN-γ的水平。结果:NKT+DC对靶细胞的杀伤作用与NKT细胞组相比,明显增强(P<0.05);NK+DC对靶细胞的杀伤作用也明显强于NK细胞组(P<0.05)。IFN-γ的表达水平,NKT+DC+靶细胞组与其他组相比最高,且在72 h时达到最高(P<0.05);NK+DC+靶细胞组IFN-γ的表达与NK+靶细胞组相比有所升高(P<0.05)。结论:DC通过刺激NKT细胞产生大量IFN-γ,有效增强NKT细胞和NK细胞杀伤白血病细胞的作用。Objective:To investigate the mechanism of the cytotoxicity of NK and NKT cells enhanced by DC from umbilical cord blood on leukemia cells.Methods:Umbilical cord blood mononuclear cells(PBMC)were extracted and cultured into DC,NK and NKT lymphocytes in vitro.The killing effects of NK,NKT,NK+DC and NKT+DC on target cells were detected by using human myeloid leukemia K-562 cells as target cells.The level of IFN-γin the co-culture supernatant was detected.Results:The killing effect of NKT+DC on target cells was significantly enhanced compared with that of NKT group(P<0.05).The killing effect of NK+DC on target cells was also significantly stronger than that of the NK group(P<0.05).The expression level of IFN-γwas the highest in the NKT+DC+target cell group compared with other groups,and reached the highest at 72 h(P<0.05).The expression of IFN-γin the NK+DC+target group was higher than that in the NK+target group(P<0.05).Conclusion:DC can effectively enhance the killing effect of NKT cells and NK cells on leukemia cells by stimulating NKT cells to produce a large amount of IFN-γ.
