摘 要:目的探讨^(125)I放射性粒子抑制裸鼠人肺腺癌移植瘤生长的机制。方法构建人肺腺癌A549荷瘤裸鼠模型,将20只裸鼠随机平均分为实验组和对照组,每组10只。实验组移植瘤植入22.2 MBq活度125I粒子,对照组植入0 MBq空源粒子。测量移植瘤长径、短径,30 d后取下移植瘤,免疫组化法检测乏氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)蛋白表达,荧光定量PCR法检测HIF-1αmRNA表达。结果实验组在植入粒子后第10天开始移植瘤生长速度放缓,30 d时体积小于对照组[(1213.8±567.6)vs(1712.1±437.4)]mm3,但差异无统计学意义(t=1.554,P>0.05)。两组移植瘤组织中HIF-1α均有表达,实验组HIF-1α蛋白表达低于对照组[(86.7±4.2)vs(102.7±3.7)],差异有统计学意义(t=9.039,P<0.05);实验组HIF-1αmRNA表达低于对照组[(0.223±0.0814)vs(0.879±0.0259)],差异有统计学意义(t=24.285,P<0.05)。结论125I粒子可抑制裸鼠人肺腺癌A549移植瘤生长,并在蛋白质、mRNA水平降低移植瘤组织HIF-1α的表达。Objective To evaluate the effect of radioactive ^(125)I seed implantation on expression of hypoxia inducible factor-1α(HIF-1α)in lung cancer A549 cell of nude mice.Methods Lung cancer xenograft models on nude mice were established,which were divided into control group and observation group randomly,with 10 mice in each group.The observation group was implanted with ^(125)I seed(radioactivity:22.2 MBq),and the control group was given blank seed(radioactivity:0 MBq).The size of xenograft tumor were detected,and the tumor inhibition rate was calculated.The expressions of HIF-1αprotein and mRNA in xenograft tumor were detected by immunohistochemical and RT-PCR.Re⁃sults When 30 days after implantation,the volume of xenograft tumor in the observation group was smaller than that in the control group,but the difference was not statistically significant[(1213.8±567.6)mm3 vs.(1712.1±437.4)mm3,t=1.55,P>0.05].The expression of HIF-1α protein of xenograft tumor in the observation group were significantly lower than that in the control group[(86.7±4.2)vs.(102.7±3.7),t=9.039,P<0.05].Compared with the control group,the expression of HIF-1α mRNA in the observation group was significantly lower[(0.223±0.0814)vs.(0.879±0.0259),t=24.285,P<0.05].Conclusion Radioactive ^(125)I seed can significantly inhibit the growth of xenograft tumor of lung cancer A549 cell in nude mice, and inhibition of HIF-1α expression may be the main mechanism.
