摘 要:目的探讨东北刺人参总皂苷改善睡眠的作用及机制。方法通过观察东北刺人参总皂苷对小鼠自主活动及翻正反射的影响,探讨其改善睡眠的作用。通过测定东北刺人参总皂苷对正常及睡眠剥夺小鼠脑部海马、皮质、下丘脑中白介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)、IL-4及IL-10含量的影响,探讨其可能的作用机制。结果与空白组相比,中剂量与高剂量东北刺人参总皂苷能显著降低小鼠自主活动次数和站立次数,可使阈剂量戊巴比妥钠所致小鼠睡眠潜伏期缩短及睡眠时间延长,还可显著增加阈下剂量戊巴比妥钠所致小鼠睡眠发生率。与空白组比较,对氯苯丙氨酸(PCPA)组小鼠脑内IL-1β、TNF-α含量明显减少,IL-4及IL-10含量升高。给予东北刺人参总皂苷后,睡眠剥夺小鼠脑内IL-1β及TNF-α含量升高,IL-4及IL-10含量降低。结论刺人参总皂苷具有镇静、改善睡眠的作用,其作用机制可能与影响脑内睡眠相关细胞因子含量有关。Objective To explore the improving effect and mechanism of Oplopanax elatus Nakai on sleep.Methods The improving effect of total saponins from Oplopanax elatus Nakai on sleep were researched through observation of the influence of the saponins on autonomous activity and righting reflex of mice.The action mechanism of the total saponins from Oplopanax elatus Nakai were researched through determination of the contents of IL-1β,TNF-α,IL-4 and IL-10 in hippocampus,cortex and hypothalamus of brains of normal and sleep deprived mice.Results Compared with the blank group,the middle and high doses of total saponins from Oplopanax elatus Nakai significantly reduced the number of autonomous activity and standing times of mice,and shortened the sleep latency and prolonged sleep time of mice caused by the threshold doses of sodium pentobarbital.It significantly increased the incidence of sleep in mice caused by pentobarbital sodium at subthreshold doses.Compared with the blank group,the content of IL-1βand TNF-αin the brain of animals in the PCPA group was significantly reduced,while the content of IL-4 and IL-10 increased.After administration of total saponins from Oplopanax elatus Nakai,the levels of IL-1βand TNF-αin the brains of sleep deprived mice increased,and the levels of IL-4 and IL-10 decreased.Conclusion The total saponins from Oplopanax elatus Nakai has sedative and sleep improving effect,and its mechanism may be related to the influence of sleep-related cytokines in brain.
