甲状旁腺素相关蛋白核定位序列及C-末端缺失对幼年小鼠下颌切牙发育影响及机制研究

摘  要:目的:研究甲状旁腺素相关蛋白(PTHrP)对2周龄小鼠下颌切牙发育的影响及作用机制。方法:分别选取2周龄PTHrP KI小鼠及同窝WT小鼠各20只,对下颌切牙行影像学、组织学、细胞凋亡、RT-PCR检测。结果:PTHrP KI组小鼠下颌切牙长度、切缘厚度、髓腔壁牙本质厚度、I型胶原阳性面积、ALP、OCN、SOD1、SOD2、CAT、GSH-Px的mRNA表达水平均明显低于WT组小鼠(P<0.01),前期牙本质比例、凋亡细胞比例均明显高于WT小鼠(P<0.01)。结论:PTHrP核定位序列和C-末端的缺失导致幼年小鼠下颌切牙细胞凋亡增加、胞外基质生成和矿化减少,导致了小鼠下颌切牙的发育异常。Objective:To study the the effects and mechanism of parathyroid hormone related protein(PTHrP)on the development of mandibular incisors in 2-weeks-old mice.Methods:The mandible and the mandibular incisors of 202-weeks-old PTHrP KI mice and 20 homozygous WT mice were examined by imaging,histology,cell apoptosis and RT-PCR.Results:The length of mandibular incisor,incisor margin thickness,dentin thickness of pulp cavity wall,positive area of type I collagen,the mRNA expression of ALP,OCN,SOD1,SOD2,CAT and GSH-Px in mice of the PTHrP KI group were significantly lower than those in WT group(P<0.01),while the ratio of predentin and the rate of apoptotic cells in PTHrP KI group were significantly higher than those in WT group(P<0.01).Conclusion:The loss of PTHrP nuclear location sequence and C-terminal may result in the increase of apoptosis of cells,decrease of extracellular matrix formation and mineralization of dentin,and the abnormal development of mandibular incisors.
