摘  要:目的探讨中药单体人参皂甙Rg3对原代培养人喉鳞癌细胞上皮-间质转化的影响及SIX1和TGF-β在上皮-间质转化中的作用。方法术中留存人喉鳞状细胞癌组织,常规处理后,体外进行人喉鳞癌原代细胞培养,待细胞传代稳定后分为对照组、顺铂组、Rg3组。顺铂组加入顺铂,使其终浓度为3μg/mL;Rg3组加入Rg3,使其终浓度为300μg/mL。处理后继续培养细胞24 h,免疫组化及Western blot检测3组细胞中SIX1、TGF-β及上皮标志物E-cadherin蛋白表达情况,RT-PCR法检测各组原代细胞中SIX1 mRNA、TGF-βmRNA及E-cadherin mRNA表达情况。结果免疫组化染色显示对照组SIX1、TGF-β蛋白强阳性表达,E-cadherin蛋白弱阳性表达;顺铂组SIX1和E-cadherin蛋白阳性表达,TGF-β蛋白强阳性表达;Rg3组SIX1蛋白弱阳性表达,E-cadherin和TGF-β蛋白强阳性表达。顺铂组及Rg3组SIX1蛋白及mRNA表达水平均明显低于对照组(P均<0.05),且Rg3组均明显低于顺铂组(P均<0.05);顺铂组及Rg3组E-cadherin蛋白及mRNA表达水平均明显高于对照组(P均<0.05),且Rg3组明显高于顺铂组(P均<0.05);3组间TGF-β蛋白及mRNA表达水平比较差异均无统计学意义(P均>0.05)。结论Rg3能够通过下调SIX1表达起到抑制人喉鳞癌细胞上皮-间质转化的作用,且其作用强于顺铂。Objective It is to investigate the effect of traditional Chinese medicine monomer ginsenoside Rg3 on the epithelial-mesenchymal transition of primary cultured human laryngeal squamous cell carcinoma cells and the role of SIX1 and TGF-βin epithelial-mesenchymal transition.Methods The human laryngeal squamous cell carcinoma tissue was preserved during the operation.After conventional treatment,the primary human laryngeal squamous cell carcinoma cells were cultured in vitro.After the cells were stable,they were divided into control group,cisplatin group,and Rg3 group.Cisplatin was added to the cisplatin group to make the final concentration 3μg/mL;the Rg3 group was added to Rg3 to make the final concentration 300μg/mL.After treatment,the cells were cultured for 24 hours.The expression of SIX1,TGF-βand epithelial marker E-cadherin protein in the cells in the three groups were detected by immunohistochemistry and Western blot,and the expression of SIX1mRNA and TGF-βmRNA and E-cadherin mRNA in the primary cells of each group were detected by RT-PCR.Results Immunohistochemical staining showed strong positive expression of SIX1 and TGF-βprotein,weak positive expression of E-cadherin protein in control group;positive expression of SIX1 and E-cadherin protein,strong positive expression of TGF-βprotein in cisplatin group;weak positive expression of SIX1 protein,strong positive expression of E-cadherin and TGF-βprotein in Rg3 group.The expression levels of SIX1 protein and mRNA in the cisplatin group and Rg3 group were significantly lower than those in the control group(P both<0.05),and the Rg3 group were significantly lower than the cisplatin group(P both<0.05);the expression levels of E-cadherin protein and mRNA in the cisplatin group and Rg3 group were significantly higher than those in the control group(all P<0.05),and the Rg3 group was significantly higher than the cisplatin group(all P<0.05);there was no significant difference in the expression levels of TGF-βprotein and mRNA among the three groups(P all>0.05)
