摘 要:为了研究亚麻油和葵花油对细鳞鲑(Brachymystax lenok)生长、脂类代谢及其肝脏脂类代谢相关基因mRNA表达的影响,试验以鱼粉、豆粕为主要蛋白源,亚麻油和葵花油为植物油源,鱼油为对照组,配制等氮等脂的3组饲料,即亚麻油组、葵花油组和鱼油组,对体重为(6.43±0.02)g细鳞鲑幼鱼进行为期63 d的饲喂试验。试验结束后,采用粗成分分析、生化分析和实时荧光定量PCR检测等方法,测定和分析了细鳞鲑生长性能、粗成分、血清生化指标和肝脏脂类代谢相关基因mRNA的相对表达量。结果表明:亚麻油和葵花油可降低细鳞鲑增重率、特定生长率,提高摄食率。亚麻油组和葵花油组细鳞鲑血清中三酰甘油含量显著高于鱼油组(P<0.05),而总胆固醇含量则显著低于鱼油组(P<0.05);亚麻油组和葵花油组细鳞鲑血清中高密度脂蛋白含量显著低于鱼油组(P<0.05),葵花油组低密度脂蛋白含量显著低于鱼油组和亚麻油组(P<0.05)。与鱼油组相比,亚麻油组和葵花油组上调了肝脏乙酰辅酶A羧化酶(acetyl-CoA carboxylase alpha,ACCα)、固醇调节元件结合蛋白-1(sterol regulatory element-binding protein-1,SREBP-1)和固醇O-酰基转移酶2 (sterol O-acyltransferase 2,SOAT2)基因mRNA表达量,下调了过氧化物酶体增殖物激活受体(peroxisome proliferation activate receptorα,PPARα)基因mRNA表达量。说明以亚麻油和葵花油替代鱼油,可降低细鳞鲑幼鱼的生长性能,调节鱼类的脂类代谢及其相关基因的表达。In order to investigate the effects of linseed oil and sunflower oil instead of fish oil on growth performance, lipid metabolism and mRNA expression of genes related to liver lipid metabolism of manchurian trout(Brachymystax lenok). Fish meal and soybean meal were the main protein sources, linseed oil and sunflower oil were vegetable oil sources, and fish oil was the control group. Three groups of feed with equal nitrogen and equal fat were prepared with these materials, namely linseed oil group, sunflower oil group and fish oil group. Young manchurian trout(6.43±0.02) g were fed for 63 days. At the end of the experiment, the growth performance indexes, crude components, serum biochemical indexes and the mRNA expression levels of genes related to liver lipid metabolism were measured by crude component analysis, biochemical analysis and real-time fluorescence quantitative PCR. The results showed that linseed oil and sunflower oil replaced fish oil reduced the weight gain, specific growth rate and feeding rate of manchurian trout. The content of triglyceride in serum of manchurian trout in linseed oil and sunflower oil group was significantly higher than that in fish oil group(P<0.05), while the total cholesterol content was significantly lower than that in fish oil group(P<0.05). The content of HDL in fish oil group was significantly higher than that in linseed oil and sunflower oil group(P<0.05). The content of LDL in sunflower oil group was significantly lower than that in fish oil group and linseed oil group(P<0.05). Compared with fish oil group, the mRNA expressions of acetyl-CoA carboxylase alpha(ACCα), sterol regulatory element-binding protein(SREBP-1) and sterol O-acyltransferase 2(SOAT2) in linseed oil and sunflower oil groups were up-regulated. The mRNA expression of peroxisome proliferation actirate receptor(PPARα) gene was down-regulated.
