摘　　要：目的评价塞来昔布联合索拉非尼对人肝癌HepG2细胞迁移与侵袭能力的影响。方法采用MTT法检测药物对HepG2细胞增殖的抑制率;通过划痕实验、Transwell实验评价药物对HepG2细胞迁移与侵袭作用的影响;采用Western blot法检测相关蛋白E-cadherin、COX-2、Notch1、Snail的表达。结果塞来昔布与索拉非尼联用有协同抑制HepG2细胞增殖的作用。与对照组比较,两药单独用药及联合用药均可抑制HepG2细胞迁移与侵袭作用(P<0.05或P<0.01)。塞来昔布无显著增强索拉非尼诱导HepG2细胞E-cadherin表达增加的作用,也无显著降低索拉非尼诱导HepG2细胞Notch1、Snail表达下降的作用;索拉非尼无显著增强塞来昔布诱导HepG2细胞COX-2表达下降的作用。结论塞来昔布可增强索拉非尼的抗HepG2细胞增殖作用,但不影响索拉非尼抑制的HepG2细胞迁移与侵袭作用。Objective To evaluate the effects of celecoxib combined with sorafenib on metastasis and invasion of human liver cancer HepG2 cells.Methods MTT assay was used to detect the inhibition rate of the drug on the proliferation of HepG2 cells;scratch test and Transwell test were used to evaluate the effects of the drug on the metastasis and invasion of HepG2 cells;Western blot was used to detect the expressions of E-cadherin,COX-2,Notch1 and Snail.Results Celecoxib combined with sorafenib can synergistically inhibit the proliferation of HepG2 cells.Compared with the control group,the metastasis and invasion of HepG2 cells were inhibited by both drugs alone and in combination(P<0.05 or P<0.01).Celecoxib did not significantly enhance the increase of E-cadherin expression induced by sorafenib in HepG2 cells,nor significantly reduce the decrease of Notch1 and Snail expression induced by sorafenib;sorafenib did not significantly enhance the decrease of COX-2 expression induced by celecoxib in HepG2 cells.Conclusion Celecoxib can enhance the anti-proliferation effect of sorafenib on HepG2 cells,but does not affect the metastasis and invasion of HepG2 cells inhibited by sorafenib.
