索氏提取装置的改良及其在烟草挥发性成分分析中的应用

摘　　要：设计了一款带固相萃取功能的索氏提取装置,并优化了其提取检测方法,应用于烟草酸性及中性挥发性成分的分析。该装置于密闭环境中加热萃取,集萃取、净化和浓缩为一体,中途不需要转移样品;而且,针对烟草样品中生物碱含量高的特点,该装置有针对性地设计了带硅胶和硅钨酸的复合固相萃取柱,在样品提取过程中即可在线去除生物碱和其他大极性成分对酸性及中性挥发性成分分析的干扰,大大简化了样品前处理操作,有效缩短了样品前处理周期。与传统的索氏提取装置及同时蒸馏萃取装置相比,带固相萃取功能的索氏提取装置能够完全吸附提取液中的烟碱,而且对烟草样品中部分重要致香成分的提取效果较理想。该装置的设计以及其配套提取方法的建立为烟草中挥发性成分的提取检测分析提供了快速可靠的方法。In this paper,a Soxhlet extractor with solid-phase extraction function was designed as well as the optimized extraction and detection method,which was used to extract and analyze the acidic and neutral volatile components in tobacco.The heating,extraction,purification and concentration were integrated and performed in a closed extractor without the need to transfer samples in the process.Furthermore,according to the characteristics of high alkaloid content in tobacco samples,the device was designed with a composite solidphase extraction column,which contained silicotungstic acid and silica gel as filler.During the sample extraction process,the alkaloids and other strong polar components could be removed online by the composite solid-phase extraction column.By using the modified Soxhlet extractor,the sample pre-processing operation was greatly simplified and the sample pre-processing cycle was effectively shortened.Compared with the traditional Soxhlet extractor and simultaneous distillation extraction device,the modified Soxhlet extractor can completely absorb nicotine in the extraction solution,and the extraction effect of some important aroma components in tobacco samples is ideal.The design of the device and the establishment of its matching extraction method provide a fast and reliable method for the extraction,detection and analysis of volatile components in tobacco.
