摘 要:目的探讨CD147在口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)中的表达情况及其对OSCC增殖作用的影响。方法收集2017年9月至2018年12月期间大连医科大学附属第一医院临床OSCC组织和癌旁组织。分别采用real-time PCR、Western blot及免疫组化方法比较CD147在OSCC组织和癌旁组织中的表达情况。人口腔鳞癌细胞UM-SCC6细胞分别转染CD147-siRNA(CD147-siRNA组)及空载体(NC组),采用CCK-8实验、集落形成实验及Ki67实验检测CD147对细胞增殖的影响。结果①real-time PCR结果显示OSCC组织中CD147基因的相对表达量高于癌旁组织(10.38±1.21 vs 1.00±0.08,P<0.05);Western blot实验结果显示OSCC组织中CD147蛋白表达量高于癌旁组织(1.27±0.15 vs 0.60±0.15,P<0.05);OSCC组织中免疫组化阳性细胞数的平均光密度值高于癌旁组织(0.43±0.06 vs 0.12±0.06,P<0.05)。②CCK-8实验测定CD147-siRNA组细胞增殖能力低于NC组(P<0.05);CD147-siRNA组细胞集落形成能力亦下降;Ki67实验平均光密度值在CD147-siRNA组中低于NC组(165.00±19.47 vs 331.33±30.09,P<0.05)。结论CD147在口腔鳞癌中高表达并可促进口腔鳞癌细胞的增殖。Objective To explore the expression of CD147 in OSCC and its effect on proliferation of OSCC.Methods OSCC tissues and normal adjacent tissues were collected from OSCC patients in First Affiliated Hospital of Dalian Medical University from September 2017 to December 2018.The CD147 expression in OSCC and adjacent tissues was detected by real-time PCR,Western blot and immunohistochemistry.UM-SCC6 cells were transfected with CD147-siRNA and NC,respectively and named as CD147-siRNA group and NC group.The effect of CD147 on the proliferation of OSCC cells was detected by CCK-8 assay,colony formation assay and Ki67 assay.Results①Real-time PCR showed CD147 relative expression was higher in OSCC tissues than in paracancerous tissues(10.38±1.21 vs 1.00±0.08,P<0.05).The protein expression of CD147 detected by Western blot was higher in OSCC tissues than in paracancerous tissues(1.27±0.15 vs 0.60±0.15,P<0.05).The IOD of immunohistochemical positive cells was also higher in OSCC tissues than in paracancerous tissues(0.43±0.06 vs 0.12±0.06,P<0.05).②The cell proliferative ability in CD147-siRNA group was lower than that in NC group by CCK-8 assay(P<0.05).The colony forming ability in CD147-siRNA group was also decreased.And Ki67 results showed that the average optical density value in CD147-siRNA group was lower than that in NC group(165.00±19.47 vs 331.33±30.09,P<0.05).Conclusion CD147 is highly expressed in oral squamous cell carcinoma,and it could promote the proliferation of OSCC.
