活血化瘀法干预下荷骨肉瘤裸鼠内环境缺氧诱导因子-1α变化机制的研究

摘　　要：目的:通过观察活血化瘀方干预骨肿瘤实验鼠模型,揭示活血化瘀法干预下荷骨肉瘤裸鼠的缺氧诱导因子(Hypoxia-inducible Factor,HIF-1α)-1α变化情况,阐明活血化瘀法的作用机制。方法:16只裸鼠造模成功后,随机分组为:A、B、C、D组,按各自给药方法灌胃,4周后处死实验鼠,采集实验鼠骨肿瘤组织及瘤体的骨髓腔组织,分别检测HIF-1α、T辅助淋巴细胞(CD4^(+)T细胞)的变化。结果:治疗后提示HIF-1α的阳性百分比变化为A组升高,B、C、D降低,其中D组最明显,依次是C组、B组,表明中药的治疗有效,联合甲氨蝶呤效果更明显,组间的比较采用方差检验,P<0.05;CD4^(+)T细胞的变化(百分比),D组的变化最明细,依次为C组、B组,组间比较P<0.05。结论:活血化瘀法对荷骨肉瘤裸鼠的治疗有效,其作用机制考虑与调节机体的HIF-1α、CD4^(+)T细胞有关。Objective:By observing the intervention of the Huoxue Huayu method(活血化瘀法)in the experimental mouse model of bone tumor,reveal the changes of HIF-1αin nude mice with osteosarcoma under the intervention of Huoxue Huayu methods,and clarify the mechanism of Huoxue Huayu methods.Methods:After the successful modeling of 16 nude mice,they were randomly divided into groups A,B,C,and D.The mice were gavage according to their respective administration methods.After 4 weeks,the mice were sacrificed,and bone tumor tissues and bone marrow of the mice were collected.Lumen tissues were tested for HIF-1αand CD4^(+)T cells changes.Results:After treatment,HIF-1αwas increased in group A,and decreased in B,C,and D.Among them,group D was the most obvious,followed by group C and group B,indicating that the TCM medicine is effective,and combined methotrexate is more effective,P<0.05;the change in CD4^(+)T cells in group D was the most detailed,followed by group C and group B,P<0.05.Conclusion:The Huoxue Huayu method is effective on nude mice bearing osteosarcoma,and its mechanism of action is considered to be related to the regulation of HIF-1αand CD4^(+)T cells in the body.
