摘 要:目的探讨移植低氧诱导因子-2α(HIF-2α)过表达的骨髓间充质干细胞(BMSCs)对脑出血(ICH)小鼠的影响.方法利用慢病毒包装质粒GV308携载HIF-2α感染BMSCs构建过表达HIF-2α的BMSCs(HIF-2α/BMSCs).参照Rynkowski法建立小鼠ICH模型,分组移植等体积的HIF-2α/BMSCs、BMSCs和生理盐水(NS)至出血部位.通过microPET-CT和Longa评分评估疗效,ELISA检测TNF-α、IL-1β、IL-6、和IL-18的血清水平,Western blot检测脑组织HIF-2α、VEGF和Ang-1蛋白的表达水平.结果与对照组相比,microPET-CT扫描显示HIF-2α/BMSCs组血肿体积显著降低(P<0.05);Longa评分显示神经功能恢复情况显著降低(P<0.05);ELISA检测显示炎症因子水平显著降低(P<0.05);Western blot检测显示血管新生因子水平显著升高(P<0.05).结论过表达HIF-2α的BMSCs可能通过抑制炎症反应、促进血管生成减轻ICH的脑血肿和改善神经功能.Objective To investigate the effect of HIF-2α-over-expressed bone mesenchymal stem cells(BMSCs)in mice after intracerebral hemorrhagic(ICH)injury.Methods HIF-2α-over-expressed BMSCs(HIF-2α/BMSCs)were constructed by transfecting BMSCs with lentiviral vector carrying HIF-2α.A mouse ICH model was established by Rynkowski method.ICH mice were randomly divided into three groups,which transplanted with the same volume of HIF-2α/BMSCs,BMSCs and physiological saline group(NS)respectively.Brain hematoma changes and neurological function scores of mice were evaluated by microPET-CT and Longa scores.Levels of tumor necrosis factor-α,interleukin-1β,interleukin-6 and interleukin-18 in mice serum were measured by ELISA method.Levels of HIF-2α,VEGF and Ang-1 protein were detected by western blot.Results Compared with NC and NS group,the volume of hematoma in the HIF-2α/BMSCs group were significantly higher(P<0.05),Longa scores were significantly reduced(P<0.05),levels of inflammatory cytokines and angiogenic factors were significantly reduced in HIF-2α/BMSCs group.Conclusions HIF-2α/BMSCs may reduce ICH cerebral hematoma and improve neurological function by inhibiting inflammatory responses or promoting angiogenesis.
