摘　　要：目的探讨子宫内膜癌组织中转移黏附蛋白(AEG-1)和相关侵袭、转移基因表达情况及其相关性。方法收集2016年7月—2018年7月在邯郸市妇幼保健院接受手术的48例子宫内膜癌患者组织标本、28例子宫内膜单纯增生组织标本和30例正常子宫内膜组织标本。使用Western印迹法测定各组织中AEG-1和基质金属蛋白酶-9(MMP-9)蛋白表达量,并分析二者表达量与子宫内膜癌患者病理特征的关系。应用RT-PCR方法检测各组织中β-连环蛋白(β-catenin)、CUL4A的mRNA表达量。Pearson分析AEG-1与MMP-9蛋白、β-catenin mRNA、CUL4A mRNA表达的相关性。结果子宫内膜癌组织中AEG-1、MMP-9蛋白表达量和β-catenin mRNA、CUL4A mRNA表达量均较子宫内膜单纯增生组织和正常子宫内膜组织中显著增高(P均<0.05)。子宫内膜癌组织中AEG-1和MMP-9蛋白表达量与患者病理分级、浸润深度及淋巴转移相关(P均<0.05),且AEG-1表达量与MMP-9蛋白、β-catenin mRNA、CUL4A mRNA表达量均呈正相关性(P均<0.05)。结论子宫内膜癌组织中AEG-1表达量异常升高,与癌细胞的侵袭和转移直接相关。Objective It is to investigate the expression of astrocyte elevated gene(AEG-1)and related invasive and metastatic gene in tissue of endometrial carcinoma(EC),and analyze the correlation between them.Methods The tissue samples of 48 patients with endometrial cancer who underwent operations,28 patients with simple endometrial hyperplasia and 30 samples of normal endometrial tissues in Handan Maternal and Child Health Care Hospital from July 2016 to July 2018 were collected.The expression of AEG-1 and matrix metalloproteinase-9(MMP-9)in the tissue of each group were detected by Western blotting,and the relationship between the expression level of the two indexes and EC pathological characteristics was analyzed.The RT-PCR method was used to detect the mRNA expression ofβ-catenin and CUL4A in each tissue.Pearson analysis was performed to analyze the correlation between AEG-1 and the expression of MMP-9 protein,b-catenin mRNA,and CUL4A mRNA.Results The expression of AEG-1,MMP-9 protein,β-catenin mRNA and CUL4A mRNA in endometrial cancer tissues were significantly higher than those in simple endometrial hyperplasia tissues and normal endometrial tissues(all P<0.05).The expression of AEG-1 and MMP-9 protein in endometrial cancer tissue was correlated with the pathological classification,depth of invasion and lymphatic metastasis of the patient(P<0.05),and the expression of AEG-1 was positively correlated with MMP-9 protein andβ-catenin mRNA,CUL4A mRNA expression(P<0.05).Conclusion The expression of AEG-1 in EC tissues is abnormally increased,which is directly related to the invasion and metastasis of cancer cells.
