摘　　要：目的探讨抑癌基因PTEN编码蛋白在癌旁胃黏膜、肠上皮化生、异型增生及胃癌组织中的表达,分析抑癌基因PTEN编码蛋白于胃癌病程中发挥的作用。方法选择2017年5月-2019年5月175例胃癌患者作为研究对象,通过免疫组化法检测癌旁胃黏膜、肠上皮化生、异型增生及胃癌组织中抑癌基因PTEN编码蛋白表达,并观察抑癌基因PTEN编码蛋白表达和胃癌病理生物学特征关系。结果癌旁胃黏膜、肠上皮化生组织内抑癌基因PTEN编码蛋白阳性表达检出率均高于异型增生、胃癌组织内抑癌基因PTEN编码蛋白阳性表达检出率,差异有统计学意义(P<0.05);淋巴结转移的胃癌组织内PTEN编码蛋白阳性表达率低于无淋巴结转移的胃癌组织内PTEN编码蛋白阳性表达率,差异有统计学意义(P<0.05);弥漫型胃癌组织内PTEN编码蛋白阳性表达率低于肠型、混合型,且肠型低于混合型,差异有统计学意义(P<0.05);管状腺癌的PTEN编码蛋白阳性表达率高于黏液腺癌、印戒细胞癌,差异有统计学意义(P<0.05).结论抑癌基因PTEN编码蛋白在癌旁胃黏膜、肠上皮化生、异型增生及胃癌组织中表达显示为进行性下调、缺失,与肿瘤细胞相关生物学行为存在相关性。Objective To investigate the expression of tumor suppressor gene PTEN protein in paracancerous gastric mucosa,intestinal metaplasia,dysplasia and gastric cancer,and to analyze the role of tumor suppressor gene PTEN protein in the course of gastric cancer.Methods A total of 175 patients with gastric cancer from May 2017 to May 2019 were selected as the subjects of this study.Immunohistochemistry was used to detect the tumor suppressor gene PTEN encoding in paracancerous gastric mucosa,intestinal metaplasia,dysplasia and gastric cancer.Protein expression,and observe the relationship between the expression of the tumor suppressor gene PTEN-encoded protein and the pathological biological characteristics of gastric cancer.Results The positive rate of PTEN-encoding protein in gastric mucosa and intestinal metaplasia was higher than those in dysplasia and PTEN protein(P<0.05).The positive rate of PTEN-encoding protein in gastric cancer tissues with lymph node metastasis was lower than that in gastric cancer tissues without lymph node metastasis(P<0.05).The positive rate of PTEN-encoding protein in diffuse gastric cancer tissues was lower than that in intestinal type and mixed type,and the intestinal type was lower than mixed type(P<0.05).The positive rate of PTEN-encoded protein in tubular adenocarcinoma was higher than that in mucinous adenocarcinoma and signet ring cell carcinoma(P<0.05).Conclusion The expression of the tumor suppressor gene PTEN protein in the gastric mucosa,intestinal metaplasia,dysplasia and gastric cancer tissues shows progressive down-regulation and deletion,which is correlated with the biological behavior of tumor cells.
