摘 要:目的探讨同时抑制血管内皮生长因子A(VEGF-A)和血管内皮生长因子C(VEGF-C)对血管瘤内皮细胞(HemECs)增殖的影响及可能的作用机制。方法采用设计的携带特异的短发夹RNA(shRNA)技术的质粒转染HemECs,将实验分为4组:空白对照组(control组)、通过shRNA技术抑制VEGF-A表达的shVEGF-A组、抑制VEGF-C表达的shVEGF-C组以及同时抑制VEGF-A和VEGF-C表达的shVEGF-A+C组。采用基因分子生物学实验手段分析各组HemECs的增殖、凋亡情况;并将HemECs种植于裸鼠,比较各组裸鼠血管瘤生长情况。结果与control组相比,VEGF-A组和VEGF-C组HemECs生长、增殖受抑制,细胞凋亡百分比升高(均P<0.05);而shVEGF-A+C组与VEGF-A组和VEGF-C组相比,HemECs的生长、增殖受抑制更明显,细胞凋亡百分比更高(均P<0.05)。裸鼠血管瘤生长情况比较,shVEGF-A组、shVEGF-C组、shVEGF-A+C组裸鼠抑瘤率分别为25.35%、49.21%、73.43%(均P<0.05),与control组相比,shVEGF-A组和shVEGF-C组肿瘤均缩小(均P<0.05),而shVEGF-A+C组比shVEGF-A组和shVEGF-C组肿瘤更小(均P<0.05)。结论VEGF-A和VEGF-C双拮抗在体内外对血管瘤内皮细胞增殖的抑制作用更为明显。Objective To explore the effect of simultaneous inhibition of vascular endothelial growth factor A(VEGF-A)and vascular endothelial growth factor C(VEGF-C)on the proliferation of hemangioma endothelial cells(HemECs)and the possible mechanism.Methods HemECs were transfected with plasmids carrying shVEGF-A(shVEGF-A group),shVEGF-C(shVEGF-C group)or shVEGF-A plus VEGF-C(shVEGF-A+C group),and the non-transfected HemECs served as control group.The proliferation and apoptosis of HemECs were measured.HemECs were transplanted in nude mice,and the growth of hemangioma in each group was compared.Results Compared with the control group,the growth and proliferation of HemECs in the VEGF-A group and the VEGF-C group were inhibited,and the percentage of apoptosis increased(all P<0.05).The growth and proliferation of HemECs in the shVEGF-A+C group were more markedly inhibited than those in the VEGF-A and VEGF-C groups,and the percentage of cell apoptosis was higher(all P<0.05).The tumor inhibition rates of nude mice in the shVEGF-A group,shVEGF-C group,and shVEGF-A+C group were 25.35%,49.21%,and 73.43%,respectively(all P<0.05).Conclusion Co-inhibition of VEGF-A and VEGF-C has more significant inhibitory effects on the proliferation of hemangioma endothelial cells in vivo and in vitro.
